摘  要:目的探讨CD45分子在γδT细胞发育过程中的作用。方法采用CD45基因敲除变种鼠,观察其Vγ3树突状表皮T细胞(Vγ3DETC),这类唯一居住于鼠类表皮组织的γδT细胞的变化。以原位免疫标记观察其表皮内Vγ3DETC的分布密度和免疫表型,以RT-PCR检测表皮细胞中Vγ3TCRmRMA的表达水平,以双荧光FCM测定Vγ3DETC在表皮细胞中及Vγ2T细胞在淋巴结细胞中所占比例,并与野生型鼠作对照。结果CD45缺失鼠表皮内Vγ3DETC的密度、免疫表型,表皮细胞Vγ3TCRmRNA表达水平以及Vγ3DETC和Vγ2T细胞分别在表皮细胞及淋巴结细胞中的比例与野生型鼠无明显差异。结论CD45分子虽在αβT细胞发育中起重要作用。Objective To investigate the role of CD45 molecule in γδT cell development. Methods The CD45 deficient mice,from which the CD45 gene had been knockout,were utilized to examine whether Vγ3 dendritic epidermal T cells (Vγ3 DETC),a subset of γδT cells uniquely residing in the murine epidermis,were altered.The density and inmmunophenotype of Vγ3 DETC were observed by in situ immunolabelling on epidermal sheets,the levels of Vγ3 TCR mRNA in epidermal cells were determined by RT PCR and the proportions of Vγ3 DETC in epidermal cells and Vγ2 T cells,another subset of γδT cells,in lymph node cells were tested by two color FCM,and all of these were compared to the wild type control mice. Results Between the CD45 deficient mice and the wild type controls,no significant differences in the dendity and immunophenotype of Vγ3 DETC,the expression of Vγ3 TCR mRNA in the epidermal cells and the proportions of Vγ3 DETC in epidermal cells and Vγ2 T cells in lymph node cells were found .Conclusions Although CD45 molecule is crucial for the development of αβT cells,this molecule is not necessary for the thymic maturation of γδT cells including Vγ3 DETC and Vγ2 T cells.
