摘  要:本实验以大脑中动脉梗塞模型(MCAO)大鼠为实验动物,以高压液相色谱仪配紫外检测技术为实验手段,在系统观察了该模型缺血后大脑海马区4种主要游离氨基酸(包括EAAs和IAAs)含量动态变化的基础上,深入研究了“醒脑开窍”针法对氨基酸类神经递质的影响,并探讨其意义。实验结果显示,脑缺血后,海马谷氨酸(Glu),天冬氨酸(Asp)和γ-氨基丁酸(GABA)含量普遍升高,而且这种异常改变主要发生在缺血早期(脑梗塞后3小时左右);针刺后Glu、Asp、Gly水平与缺血组比较显著降低(P<0.01),而GABA含量则明显升高(P<0.01)。研究结果表明,氨基酸类递质的兴奋性神经毒性,在缺血性脑损伤过程中,特别是在缺血早期,可能起着重要的病理作用,而针刺能够减轻该兴奋性神经毒性,考虑这可能是针刺治疗脑梗塞的重要机制之一。The middle cerebral artery occlusion (MCAO) model rat was used as an animal pattern in the experiment. The experimental methods were high pressure liquid chromatography and ultraviolt-spectrophotometry. On the base of observating the dynamic change of four kinds of main free amino acids content in hippocampus, the effects of 'Xing Nao Kai Qiao' acupuncture method on the transmitter of amino acids were researched deeply. The experimental result showed: the content of glutarnic acid (GLU), aspartic acid (ASP), glycine (GLY) and γ-aminobutyric acid (GABA) increased after cerebral infarction. This abnormal change often appeared in early infarction (about three hours after cerebral infarction); as compared with the group of infarction, the levels of GLU, ASP, GLY after acupuncture were reduced markedly (P <0. 01 ), and the content of GABA was increased (P < 0. 01 ).The experimental result indicated that the excitatory neurotoxicity of the transmitter of amino acids may play an important pathological role in ischemic cerebral lesion, especially in early ischemia. Acupuncture could reduce excitatory neurotoxicity. This might be one of the major mechanisms of acupuncture treatment on cerebral ischemia.
