摘　　要：目的探讨姜黄素诱导视网膜母细胞瘤(RB)细胞株Hxo-rb44细胞DNA损伤和周期时相变化的关系。方法10～50μmol/L姜黄素作用6～24h后,MTT比色法检测细胞生长活性,流式细胞仪测定细胞周期时相变化,彗星实验检测细胞DNA损伤。结果姜黄素能显著诱导RB细胞株Hxo-rb44细胞DNA损伤,Olive尾矩与剂量正相关,细胞周期阻滞于G0/G1期。结论姜黄素通过诱导RB细胞株Hxo-rb44细胞DNA损伤,可使细胞周期时相改变。Objective To investigate the effect of curcuma on DNA injury and cell cycle of retinoblastoma cell Hxo-rb44. Methods Hxo-rb44 cells were treated with 10 to 50ummol/L curcuma 6 to 24 hours. Cell activity was measured by MTT assay. Changes of cell cycle were detected using flow cytometry. DNA injury was evaluated by comet assay.Results Curcuma induced DNA injury of retinoblastoma cell Hxo-rb44 significantly. A positive correlation was observed between Olive tail moment value and dosage. Cell cycle was blocked at G0/G1 stage. Conclusion Curcuma can induce DNA injury and inhibit cell cycle of retinoblastoma cell Hxo-rb44.
