HTLV-I相关抗原及其抗体在AML细胞表面的表达

摘　　要：应用直接免疫金银染色法和直接酶免疫法分别检测３７例急性髓系白血病（ＡＭＬ）患者白血病细胞培养前后其表面ＨＴＬＶ－Ｉ相关抗原及其抗体的变化。结果表明，培养前ＨＴＬＶ－Ｉ相关抗原的患者阳性率、细胞阳性率（％，ｘ±ｓ）分别为２７．０％（１０／３７）、１６．８９±２７．７４，培养６天后分别上升至７８．４％（２９／３７）、４３．４４±３０．０６，分别经χ２检验和ｔ检验表明均有显著性差异（Ｐ＜０．０１，Ｐ＜０．００１）。培养前ＨＴＬＶ－Ｉ相关抗体的患者阳性率、细胞阳性率（％，ｘ±ｓ）分别为７３．０％（２７／３７）、４８．２１±３４．３０，培养６天后未能检测出ＨＴＬＶ－Ｉ相关抗体。２０例对照采用相同的细胞分离、培养和检测手段无阳性发现。进一步分析表明，培养前ＡＭＬ细胞表面ＨＴＬＶ－Ｉ相关抗原及其抗体表达的细胞阳性率之间呈明显负相关（ｒ＝－０．５６，Ｐ＜０．０１）。从而推论ＡＭＬ可能与外源性。Specific retroviral antigens and related antibodies on leukemic cells from 37 patients with AMLs were detected by direct immune gold silver staining and direct enzyme immunoassay,respectively.The results showed that the positive rates and mean positive cell percentages of retroviral antigens before cultrue were 27.0%,16.9%,and after culture were 78.4% 43.4%,respectively with a statistically significant difference (χ 2, P <0.01,t test P <0.001),respectively.On the other hand,the positive rates and mean positive cell percentages of retroviral antibodies before cultrue were 73.0%,43.2%,respectively,and after culture were not detected.Retroviral antigens and antibodies were not found on the surfaces of lymphocytes from 20 control's.Correlation analyses suggested that the positive cell percentages of retroviral antigens were negatively correlated with that of antibodies ( r =-0.56, P <0.01).These data indicated that AML may be related to infection of exogenous retrovirus partially homologous to HTLV Ⅰ.
