摘 要:目的对当归六黄汤分煎样品与合煎样品中阿魏酸、盐酸小檗碱、黄芩苷的含量进行比较,从而为其临床应用提供依据。方法均采用反相液相色谱法,DIKMA ODS-C18柱(5μm 4.6 mm×250 mm),以0.085%磷酸-乙腈为流动相梯度洗脱,流速为1.0 ml.min-1,柱温:35℃,检测波长为326nm测定阿魏酸。以乙腈-0.5%的三乙胺(磷酸调pH=3.00)(27∶73)为流动相,流速为1.0 ml.min-1,柱温:30℃,检测波长为343 nm测定盐酸小檗碱。以甲醇-水-磷酸(55∶45∶0.2)为流动相,检测波长为280 nm,流速为1.0ml.min-1,柱温30℃测定黄芩苷。结果3个指标性成分在分煎样品中平均含量均大于合煎样品中平均含量。结论从中药复方汤剂中指标性成分的含量考虑,当归六黄汤配方颗粒应用于临床具有一定的可行性。Objective To compare the contents of ferulic acid,berberine ,baicalin in single medicine decoction and mixed medicine decoction of Danguiliuhuang decoction, and to provide a scientific basis of Danguiliuhuangdecoction for clinic. Methods The samples were determined by HPLC. DIKMA ODS-C18 column (5 μm, 4.6 mm ×250 mm)was used, the mobile phase consisted of 0. 085% phosphorie-acetonitrile with flow rate 1.0ml · min^-1, The detection wavelength was set at 326 nm and the column temparature was 35℃ and Ferulic Acid was detected. The mobile phase consisted of acetonitrile and water(27:73 ) , which was adjusted to pH3.0 by 0.5% triethylamine solution and pbosphofic acid with flow rate 1.0 ml · min^-1 , the detection wavelength was seted at 343nm ,the column temparature was 30℃ and berberine was detected . The mobile phase consisted of methanol-water-phosphoric (55:45:0.2)with flow rate 1.0 ml · min^-1. The detection wavelength was set at 280 nm,the column temprature was 30℃ and baicalin was detected. Results The contents of ferulic acid ,berberine ,baicalin in single medicine decoction was higher than in mixed medicine decoction. Conclusion It can be used in clinic as a reference.
