摘 要:目的检测在心肌缺血再灌注损伤中TNF的表达,并初步探讨其机制。方法采用Langendorff模型及大鼠离体心脏灌注模型,心肌组织缺血30 min后、再灌注30、60、120 min时,用原位杂交、免疫组化和酶联免疫法检测心肌细胞中TNF mRNA、蛋白的表达。结果缺血再灌注实验组心肌组织中的TNF蛋白水平比对照组明显升高;实验组心肌细胞中的TNF mRNA和蛋白表达明显,染色呈强阳性或极强阳性(t=2.689,P<0.05),并且呈阳性表达的细胞是心肌细胞。结论缺血再灌注可刺激心肌细胞大量表达TNF,并且心肌组织中的TNF主要由心肌细胞表达产生。Objective To observe the expression of TNF in rats with myocardial isehemic reperfusion injury, and explore the biological characteristics of it. Methods Adult male Wistar rats were subjected to myocardial isehemie reperfusion, after the myocardial tissue isehemic 30 min, with reperfusion for 30 rain, 60 min, and 120 rain. The mRNA and protein expressions of TNF in myocardial cells were detected by in sire hybridization, immunohistochemistry and ELESA. Results The mRNA and protein expressions of TNF in myocardial cells were higher in the myocardial ischemic reperfusion group than in the control group, t = 2. 689, P 〈 0.05, and the most expression of TNF was in myocardial cells. Conclusions The isehemic reperfusion could stimulate myocardial cells expressing TNF, and most TNF is expressed in myocardial cells.
