摘  要:[目的]获取性状优良的聚羟基烷酸(PHAs)合成菌。[方法]以东湖底泥、山坡裸地土、肥沃菜园土和油脂浸润土为初始样品,经富集、分离纯化、苏丹黑B染色、红外光谱及气相色谱定性定量分析筛选出具PHAs合成能力的细菌菌株。[结果]苏丹黑B染色结果表明,在分离出的12株细菌中有3株细菌具有PHAs合成能力,其编号分别是Zj5、Zj8和Zj9。3株细菌均在1 741 cm-1附近呈现吸收峰,菌株Zj8和Zj9的红外谱图相似。菌株Zj5、Zj8和Zj9的PHAs含量分别为146.85、0.65和3.48 mg/L,菌株Zj5被初步鉴定为假单胞菌属细菌。菌株Zj5在pH值为5.0-9.0的环境中均能生长,其生长的最适pH值为7.6;在25-35 ℃下均能正常生长,其最适培养温度和通气量分别为30 ℃和100 ml。[结论] 菌株Zj5、Zj8和Zj9有PHAs合成能力且菌株Zj5的合成能力最强。[ Objective ] The purpose of the research was to obtain bacterial polyhydroxyalkanoic acids (PHAs) producers with good characters. [ Method] With bottom sludge from Donghu Lake, bare soil from hillside, fertile vegetable soil and oil-soaked soil as initial samples, the bacterial strains with ability of synthesizing PHAs were screened out through enriching, isolation and purification, staining with Sudan black B and qualitative and quantitative analyses by infrared spectroscopy and gas chromatography. [ Result ] The results of staining with Sudan black B showed that there were 3 strains with ability of synthesizing PHAs among the 12 isolated strains and their numbers were Zj-5, Zj-8 and Zj-9 resp. All the 3 strains took on adsorption peaks at the points near 1 741 cm-1 and the infrared spectrums of the strains Zj-8 and Zj-9 were similax. The PHAs contents of the strains Zj-5, Zj-8 and Zj-9 were 146.85, 0. 65 and 3.48 mg/L and the strain Zj-5 was identified as Pseudomonas sp preliminarily. The strain Zj-5 could grow in the environments with pH value in 5 -9 and the optimum pH value for its growth was 7.6; it could grow at the temperature of 25 -35 ℃ normally and the optimum culture temperature and ventilation volume for its growth were 30℃ nd 100 ml resp. [ Conclusion] The strains Zj-5, Zj-8 and Zj-9 had ability of synthesizing PHAs and the synthesis ability of the strains Zj-5 was strongest.
