摘 要:[目的]对河豚4SNc-Tudor蛋白(SN4TDR)进行序列与结构相关的生物信息分析,为认识该蛋白结构与功能提供借鉴。[方法]应用双向电泳及基质辅助激光解吸离子化-飞行时间质谱(MALD I-TOFMS)技术,从河豚肝脏总蛋白中鉴定到河豚的4SNc-Tudor结构域蛋白。并对其进行生物信息学分析。[结果]首次从河豚肝脏总蛋白中鉴定到河豚的4SNc-Tudor结构域蛋白SN4TDR。河豚SN4TDR定位于细胞质,无信号肽,为非分泌蛋白,有多个磷酸化位点。蛋白SN4TDR存在α-螺旋、β-折叠和无规则卷曲结构,有一个可能形成跨膜结构的片段。并用同源建模的方法,预测了河豚SN4TDR的三维结构模型。[结论]分析河豚SN4TDR基因及蛋白结构,有助于进一步研究该蛋白及其基因的分子和生物学功能。[Objective ] To provide a structural and functional bioinformatics analysis of 4SNc-Tudor domain protein (SN4TDR) in Takifugu rubripes, which can be refered to subsequently. [ Methods ] Two-dimensional gel electrophoresis (2-DE) and matrix-assisted laser desorption/ionization time of flight mass spectrometry( MALDI-TOF MS) were used to identify the SN4TDR protein from T. rubripes liver. Various methods were used in bioinformatics analysis. [ Results] The SN4TDR protein was first identified in T. rubripes. And it's a non-secreted protein with multiple phosphorylation sites and without signal peptide, and located in the cytoplasm. The SN4TDR protein contains such structures as αhe- lix, [5-sheet, random coil and a transmembrane structure. A 3D model of SN4TDR was built by using the homology modeling, [ Conclusion ] The bioinformaties analysis of SN4TDR would be engaged to the comprehension of molecular and biological functions of this protein.
