摘  要:目的探讨原花青素(procyanidin,PC)对2型糖尿病局灶性脑缺血大鼠脑组织含水量和脑梗死面积的影响。方法将60只大鼠随机分为正常对照组、2型糖尿病单纯缺血组、PC低剂量组(50 mg/kg)、中剂量组(100 mg/kg)、高剂量组(200 mg/kg)。除正常对照组外,其余各组大鼠高脂高糖饲料喂养4周后,小剂量链脲佐菌素(Streptozotocin,STZ)一次性尾静脉注射,制备2型糖尿病大鼠模型。局灶性脑缺血24 h后处死大鼠,计算脑含水量和脑梗死面积。结果与正常对照组比较,2型糖尿病单纯缺血组大鼠脑组织的含水量和梗死面积明显增加(P<0.01);而PC各组均能不同程度的减少2型糖尿病局灶性脑缺血大鼠脑组织含水量和减小梗死面积(P<0.05 vs P<0.01),且以中、高剂量组更为明显,但PC中、高剂量组间差异无显著性(P>0.05)。结论PC可减轻脑水肿程度、缩小局灶性脑缺血大鼠脑梗死面积,具有神经保护作用。Objective To investigate the effect of procyanidin(PC) on content of water in brain and area of cerebral infarction in type 2 diabetes mellitus SD with focal cerebral ischemia.Methods The SD 60 rats were divided randomly into normal group,model group,low-dose PC group,mid-dose PC group and high-dose PC group.Feeding fats with glucose forage for four weeks,STZ was injected lump-sum through vena caules,in order to make NIDDM rats.24 hours after MCAO,content of water in brain and area of cerebral infarction was calculated.Results Comparing with normal group,the content of water in brain and area of cerebral infarction in model group was increased significantly(P0.01),however,it decreased in different PC groups especially in mid-dose PC group and high-dose PC group(P0.05vs P0.01).But there was no significant differences between them(P0.05).Conclusions Procyanidin can reduce the area of cerebral infarction in focal ischemic rats,reduce the level of cerebral edema and has an effectively neuroprotective effect.
