摘 要:目的观察抗脑抗体对脑胶质瘤细胞C6的增殖、凋亡和侵袭力的影响。方法将C6细胞分为实验组和对照组,实验组加入不同浓度的抗脑抗体,对照组不加抗脑抗体;分别采用MTT法、流式细胞术及Boyden小室测算C6增殖抑制率、细胞凋亡率及穿膜细胞数。结果实验组加抗脑抗体5、10、20、30、40μg/ml处理后,C6增殖抑制率分别为16.3%±4.4%、18.7%±3.2%、22.6%±3.0%、27.7%±1.4%、39.9%±3.3%,对照组为0;实验组加抗脑抗体5、40μg/ml处理后,C6细胞凋亡率分别为17.257%±5.964%、28.677%±6.748%,对照组为6.043%±0.421%;实验组加抗脑抗体5、20、30μg/ml处理后,C6穿膜细胞数分别为(3 266.67±251.66)、(7 133.33±305.51)、(15 000.00±2 000.00)个,对照组为(1 533.33±152.75)个。实验组各浓度间相比,P均<0.05;与对照组相比,P均<0.05。结论抗脑抗体可抑制脑胶质瘤细胞的增殖、侵袭,并诱导其凋亡。Objective To observe the effect of antibrain-antibody(AB-Ab) on cell proliferation,apoptosis and invasive ability of C6 glioma in vitro. Methods C6 glioma cells were divided into experiment group and control group. Different concentrations of AB-Ab were administrated to the experimental groups but the control group. The proliferation inhibition rate,apoptosis rate and the trans-membrane cell numbers of C6 cells were measured with respectively MTT, flow eytometry and Boyden chamber. Results The C6 cell proliferation inhibition rates were 16.3%±4.4%、18.7%±3.2%、22.6%±3.0%、27.7%±1.4%、39.9%±3.3%,0, in 5,10,20,30,40 p.g/ml AB-Ab groups and control group respectively. The experimental groups received AB-Ab 5,40 μg/ml. The C6 cell apoptosis rates were 17. 257%±5. 964%, 28. 677%±6.748% , 6. 043%±0.421%, in 5,40 μg/ml AB-Ab groups and control group respectively. The trans-membrane cell numbers were 3 266.67±251.66, 7 133.33±305.51, 15 000.00±2 000.00, 1 533.33±152.75,in 5,20,30μg/ml AB-Ab groups and control group respectively. The above-mentioned index had obvious statistically significant difference in different experiment groups and control group ( P 〈 0.05 ). Conclusion AB-Ab can inhibit proliferation, invasive ability and induce apoptosis of C6 glioma cells.
