摘  要:目的观察cripto-1基因小干扰RNA(siRNA)对乳腺癌细胞侵袭力的影响。方法将人乳腺癌MDA-MB468细胞分为4组,A组不处理,B组转染空载脂质体,C组转染错配siRNA,D组转染分别转染含3.125、6.25、12.5 nmol/L cripto-1基因siRNA的脂质体。分别于处理24、48、72 h收集各组细胞。采用实时定量RT-PCR法和Western blot法检测MDA-MB468的cripto-1 mRNA和蛋白,采用软琼脂集落培养试验和Boyden小室检测MDA-MB468的锚着不依赖性增殖和侵袭能力。结果与A、B、C组比较,D组MDA-MB468的cripto-1 mRNA、cripto-1蛋白、集落形成数量、穿膜细胞数均呈浓度依赖性减少(P均<0.005)。结论cripto-1基因siRNA可抑制乳腺癌细胞侵袭。Objective Toinvestigute the effect of cripto-1 small interfering RNA(siRNA) on invasion of human breast cancer cell. Methods After human breast cancer ceil line MDA-MB468 were transfected by cripto-1 siRNA, the mRNA and protein of eripto-1 gene were detected by real time RT-PCR and Western blot assay, respectively. The anchorage-independent growth was exmined by colon formation in soft agar,and invasion ability was evaluated by boyden chamber model. Results The siRNA could down-regulate the level of mRNA and protein of cripto-1 in a dose- and time-dependent manner. Suppression of of cripto-1 expression can inhibit anchorage-independent growth in a dose-dependent manner, and invasion ability of breast cancer cell. Conclusion Cripto-1 siRNA eand inhibit invasion ability of human breast cancer cell.
