摘  要:目的研究左旋四氢巴马汀(l-THP)对氯化钾所致大鼠海马神经元胞内钙超载的影响。方法原代培养大鼠海马神经元细胞,分为4组:空白对照组、l-THP1μmol/L组、l-THP10μmol/L组、l-THP100μmol/L组。应用激光扫描共聚焦显微镜实时扫描,观测大鼠海马神经元胞内钙的荧光强度变化。结果在静息状态下(1~100)μmol/L左旋四氢巴马汀对胞内钙浓度([Ca2+]i)均无影响。左旋四氢巴马汀对60mmol/LKCl诱导的外钙内流引起的胞浆钙升高有抑制作用。对照组、1μmol/Ll-THP、10μmol/Ll-THP、100μmol/Ll-THP的峰值荧光强度增加分别为(64.3±2.8)%、(46.3±3.1)%、(34.2±1.5)%、(20.8±1.2)%(P<0.05)。左旋四氢巴马汀对10mmol/LCaffeine诱导细胞内钙库的钙释放,引起的胞浆钙升高有抑制作用。对照组、lμmol/Ll-THP、10μmol/Ll-THP、100μmol/Ll-THP峰值荧光强度增加分别为(53.9±4.3)%、(48.2±3.9)%、(44.3±4.6)%、(35.5±3.2)%(P<0.05)。结论左旋四氢巴马汀可以通过减轻损伤诱发的海马神经元细胞内钙超载程度来保护海马神经元。Objective To study the effects of L- Tetrahydropalmatine(1 THP) on KC1 induced calcium overloading injury in cultured hippocampal neurons. Methods Hippocampal neurons were obtained from newborn Wistar rats and randomly divided into four groups:controlgroup.l-THP 1 μmol/L group,1-THP 10μmol/L group and 1-THP 100 μmol/L group. They were loaded with Ca2+ sensitive fluorescent indicator Fluo- 3/AM. Ca2+ concentration([Ca2+]i) represented by fluorescent intensity was measured by laser scanning confocal microscope. Results At resting status, [Ca2- ]i was not affected by I. - Tetrahydropalmatine (1 - 100 μmol/L). I.- Tetrahydropalmatine at the concentration of 1- 100 μmol/L significantly reduced the [Ca2+ ]i which induced by 60 mmol/L KCl, The fluorescent intensity ([Ca2+]i) were increased (64.3±2.8)% ,(46.3±3.1)G,(34.2±1.5)G,(20.8±1.2)G in control,1-THP 1,10 and 100 fLmol/I.,respectively(P〈0. 05). L - Tetrahydropalmatine at the concentration of 1 - 100 μmol/L significantly reduced the [Ca2+ ]i which induced by 10 retool/I. Caffeine. In Control, 1 - THP 1,10 and 100μmol/L, the fluorescent intensity ([Ca2+] ]i) were increased (53.9±4.3)% ,48.2±3.9)% ,(44.3±4.6)%, (35.5±3.2)% ,respectively(n=6,P〈0.05). Conclusion l Tetrahydropalmatine could alleviate injury induced calcium overloading injury in cultured hippocampal neurons.
