摘 要:目的观察RhoA和Rho激酶Ⅰ(ROCKⅠ)蛋白在前列腺癌中的表达,探讨RhoA/ROCKⅠ信号传导途径在前列腺癌形成过程中的可能作用。方法选取2005年1月-2008年5月住院手术的前列腺癌患者20例(前列腺癌组),年龄57~86(73±7)岁,行根治性前列腺切除术并留取前列腺癌组织,以及良性前列腺增生患者36例(良性前列腺增生组),年龄50~83(71±7)岁,行尿道前列腺电切除术,留取增生的前列腺组织。免疫组化法测定两组前列腺癌与增生前列腺组织中RhoA和ROCKⅠ蛋白的表达。结果前列腺癌组RhoA和ROCKⅠ蛋白的表达水平(0.225±0.068、0.273±0.070)均明显高于良性前列腺增生组(0.126±0.047、0.176±0.040,P<0.01)。相关分析显示,RhoA与ROCKⅠ蛋白的表达水平呈显著正相关(r=0.487,P<0.01)。结论RhoA和ROCKⅠ蛋白在前列腺癌中表达增加,且高于良性前列腺增生组织;RhoA/ROCKⅠ信号传导途径可能参与了前列腺癌的形成。Objective To detect the expression levels of RhoA and Rho kinaseⅠ(ROCKⅠ)proteins in prostate carcinoma,and approach the possible mechanism of RhoA/ROCKⅠsignal transduction system in the pathogenesis of prostate carcinoma.Methods Tissues from 20 patients with prostate carcinoma(prostate carcinoma group)aged from 57 to 86 were obtained by radical prostatectomy,and that from 36 cases of benign prostatic hyperplasia(benign prostatic hyperplasia group)were obtained by transurethral resection of prostate(TURP)in patients aged from 50 to 83.The diagnosis of all the patients was confirmed by pathological examination after operation from Jan.2005 to May 2008.The expression levels of RhoA and ROCKⅠ proteins in tissues of prostate carcinoma and benign prostatic hyperplasia were detected by immunohistochemical methods.Results The expression levels of RhoA and ROCKⅠ proteins in prostate carcinoma group were significantly higher than those in benign prostatic hyperplasia group(0.225±0.068 vs 0.126±0.047,0.273±0.070 vs 0.176±0.040,P〈0.01),respectively.There was significant positive correlation of expression levels between RhoA and ROCKⅠ proteins(r=0.487,P〈0.01).Conclusions Expression levels of RhoA and ROCKⅠ proteins are increased in prostate carcinoma,and higher than those in benign prostatic hyperplasia tissue.RhoA/ROCKⅠ signal transduction system may be involved in the development of prostate carcinoma.
