芦荟大黄素、阿霉素联合应用对肝癌HepG2细胞增殖的影响

摘　　要：目的体外观察芦荟大黄素(AE)、阿霉素(ADM)单用及联合应用对人肝癌HepG2细胞增殖的影响。方法采用MTT法观察不同浓度AE、ADM单独和联合应用对HepG2细胞生长的抑制作用,并计算两药相互作用系数(CDI),检验两药联合协同性。结果两药单独作用48 h对HepG2细胞的抑制率,随着药物浓度的增加均相应增加,呈剂量依赖性。两药联合作用48 h对HepG2细胞的抑制率,较同剂量单独用药亦有相应增加;CDI分析显示两药联用在相应浓度下具有协同性,而且两个联合用药组表现为协同性显著。结论芦荟大黄素、阿霉素单用及联用对人肝癌HepG2细胞有增殖抑制的作用,且两药联合具有协同性。Objective To study the effect of the proliferation about HepG2 cells were treated by aloe-emodin,doxorubicin alone and both of them in vitro.Methods Using different concentrations of AE,ADM alone and in combination on the growth inhibition of HepG2 cells by MTT assay,and Calculation the coefficient of drug in interaction,testing two-drug combination of synergistic.Results when treated with aloe-emodin or doxorubicin for 48h,with the increase of drug concentration,the growth inhibition rate increased.And 48h of the two drugs combined effects of inhibition rates of HepG2 cells compared with the same dose of medication is also a corresponding increase in a separate,CDI analysis shows that the two drugs combined with the corresponding concentration of synergistic nature,and there are two significant synergies.Conclusion Aloe-emodin,adriamycin alone and combined with human hepatoma HepG2 cells proliferation inhibition,and the two-drug combination has a synergy.
