摘  要:目的探讨肿瘤坏死因子相关凋亡诱导配体(TRAIL)联合阿霉素和IFN-γ应用对人骨肉瘤细胞生长抑制和诱导凋亡的作用。方法用MTT测定TRAIL、阿霉素、IFN-γ单独或联合应用对人骨肉瘤细胞MG-63的体外增殖抑制作用,并检测各组细胞凋亡率。结果TRAIL、阿霉素、IFN-γ三者联合作用于MG-63细胞后,能显著增强对MG-63的杀伤、抑制增殖及诱导凋亡作用,明显高于单独应用TRAIL组或阿霉素组(P<0.05)。结论TRAIL、阿霉素、IFN-γ三者联用能显著提高骨肉瘤细胞MG-63生长抑制和诱导凋亡作用。Objective To investigate the synergistic induction of cytotoxicity and apoptosis by the combination of tumor necrosis factor related apoptosis inducing ligand (TRAIL), adriamycin(ADM) and IFN-γ in osteosarcoma MG-63 cells. Methods MTT assays were used to evaluate the cytotoxic effects of MG-63, and flow cytometric analyses were used for the assessment of apoptosis rate after different treatments. Results The combined use of the three treatments induced significantly higher apoptosis and cytotoxicity than exposure to TRAIL, ADM or IFN -γ alone, but the combination of two of the three agents did not show synergistic effects. Conclusion The combination of TRAIL, ADM and IFN-γ could significantly increase cytotoxic and apoptosis effects on MG-63 cells.
