胃腺癌组织中NKG2D及DAP10表达的初步研究

摘  要:目的观察NKG2D、DAP10在胃腺癌和癌旁正常组织中的表达情况,探讨两者在胃癌发生发展中的可能作用。方法对20例胃腺癌患者的胃癌及癌旁正常组织,应用免疫组化SP法检测NKG2D、DAP10的表达水平及差异,分析它们在不同组织中的表达率、表达强度及相互关系。结果胃癌组织中NKG2D的表达率明显低于癌旁正常组织(55%vs100%,P<0.01);且胃癌组织中NKG2D的表达强度显著低于癌旁正常组织(P<0.01)。胃癌组织中DAP10的表达率虽然高于癌旁正常组织,但差异无显著性意义(100%vs90%,P>0.05);其表达强度明显强于癌旁正常组织(P<0.01)。胃癌组织中NKG2D与DAP10的表达强度无明显相关性(r=-0.425,P>0.05)。结论胃癌组织中NKG2D的表达下调,而DAP10的表达上调,提示胃癌组织中NKG2D/DAP10信号途径存在明显紊乱,可能与胃癌免疫逃逸有关。Objective To investigate the protein expression levels of both NKG2D and DAP10 in gastric adenocarcinoma and pericancerous normal tissues,and to research the relationship between their expression and development of gastric cancer.Methods The expressions of NKG2D and DAP10 protein were detected by immunohistochemistry in 20 gastric adenocarcinoma and its pericancerous normal tissues. Then we analyzed the expression rate and intensity of NKG2D and DAP10 in different tissues,and analyzed the relationship between their expression.Results The positive expression rate and expression intensity of NKG2D protein in gastric adenocarcinoma lower than that in pericancerous normal tissues (P0.01).The expression intensity of DAP10 in gastric adenocarcinoma was higher than that in pericancerous normal tissues (P0.01),but not positive expression rate of DAP10 (P0.05).There was no correlation between NKG2D and DAP10 expression in gastric cancer tissue (r=-0.425,P0.05).Conclusion The NKG2D expression downregulated in gastric cancer,but DAP10 expression upregulated.There was significant confused about NKG2D/DAP10 signal path in gastric cancer tissues,which may take part in immune escape of gastric cancer.
