摘  要:目的观察心脉龙对缺血再灌注损伤的幼兔心肌有无保护作用,探讨其对未成熟心肌的作用机制。方法54只3周龄左右幼兔随机分为假手术(Sham)组、缺血再灌注(I/R)组和心脉龙(XML)组,每组18只。制备幼兔心肌缺血再灌注损伤模型,取心肌组织制作心肌组织匀浆和石蜡切片(HE染色),观察一氧化氮合酶(NOS)、丙二醛(MDA)在各组幼兔心肌组织表达的动态变化。结果①XML组内皮型NOS(ecNOS)活性降低幅度显著低于I/R组(P<0.01)。②再灌注后心肌组织的MDA显著升高(P<0.01),XML组MDA含量显著低于I/R组(P<0.01)。③再灌注后XML组肌酸磷酸激酶(CK)值升高幅度显著低于I/R组(P<0.01)。④病理切片显示心脉龙使再灌注后炎症细胞的浸润明显减少,心肌细胞变性坏死程度减轻。结论心脉龙通过抗脂质过氧化作用,减少氧自由基的产生,保护冠状动脉内皮,增加内源性一氧化氮(NO)的产生,减轻再灌注造成的未成熟心肌组织结构的损伤和功能障碍。Objective To observe if the Xinmailong has the protection in immature rabbit myocardial ischemia reperfusion,and explore the mechanism.Methods fifty-four immature rabbits were random divided into three treated group:Sham group(n=18),ischemia reperfusion(I/R)group(n=18) and Xinmailong(XML) group(n=18).By the experimental model of anesthetized immature rabbit MIRI,the effect of Xinmailong on the content of MDA,NOS,myocardial enzyme and the myocardial fine structural were observed.Results ①The decreased argument of the activity of ecNOS in XML group significient lower than the I/R group(P0.01).②Compared with sham group,the content of MDA in I/R group and XML group were all evidently increased(P0.01).Compared with I/R group,the Xinmailong could evidently decrease the content of MDA(P0.01).③At every reperfusion time point,the increased argument of CK in XML group was lower than I/R group(P0.01).④Xinmailong could inhibit the infiltration of inflammatory cell,lighten the myocardial cell degeneration and necrosis.Conclusion The protective effects of Xinmailong maybe that inhibit lipid peroxidation,decrease the production of oxygen free radical,and protect the endothelial of coronary artery,and increase the content of nitric oxide,and lighten the structure injury and dysfunction.
