摘  要:目的探讨新型酞菁类光敏剂锌酞菁H1(ZnPcH1)介导的光动力疗法(ZnPcH1-PDT)对鼻咽癌CNE1细胞的杀伤作用。方法采用人鼻咽癌CNE1细胞作为研究对象,应用MTT比色法和细胞集落形成实验检测PDT对CNE1细胞增殖的影响,采用AO/EB荧光染色法、TUNEL、DNA倍体分析检测细胞的死亡方式。结果MTT比色法及细胞集落形成实验结果显示,ZnPcH1-PDT可抑制鼻咽癌CNE1细胞增殖抑制率,并随ZnPcH1的增加而增高(P<0.05);AO/EB荧光染色法、TUNNEL、DNA倍体分析结果均表明ZnPcH1-PDT能够诱导CNE1细胞凋亡,凋亡率呈时间依赖性。结论ZnPcH1-PDT能够有效地抑制鼻咽癌CNE1细胞增殖,诱导该细胞凋亡,拥有良好的应用前景。Objective To investigate the killing effect of ZnPcH1-PDT on the nasopharyngeal carcinoma cells. Methods The human nasopharyngeal carcinoma cell line CNE1 was selected to research.The survival rate of cells was assessed by MTT method and colony formation,the death model was detected by AO/EB,TUNEL and cell cycle test. Results The results of MTT method and colony formation showed that ZnPcH1-PDT both had significant killing effect on CNE1 cell.The results of AO/EB,TUNEL and cell cycle test showed that ZnPcH1-PDT could induce apoptosis effectively.The apoptosis rate was increased in a time-dependent manner. Conclusions ZnPcH1-PDT can inhibit nasopharyngeal carcinoma cells proliferation and induce apoptosis in a time-dependment manner.
