摘 要:目的:利用蛋白质组学的研究方法,找出增生性瘢痕、瘢痕疙瘩与正常皮肤组织成纤维细胞的差异蛋白质,为防治病理性瘢痕提供新思路.方法:应用2-DE技术建立三种组织细胞的蛋白质图谱,分析后找到差异蛋白点,用质谱分析仪及数据库确定蛋白质的种类.结果:选择增生性瘢痕、瘢痕疙瘩和正常皮肤表达的差异蛋白9个,鉴定出差异蛋白5个,分别为:磷脂酰胆碱结合蛋白、银屑病型脂肪酸结合蛋白、Ⅰ型胶原蛋白、Ⅲ型前胶原蛋白、Ⅰ型前胶原蛋白.结论:共找出增生性瘢痕、瘢痕疙瘩与正常皮肤间5种差异蛋白,为研究病理性瘢痕的发病机制提供了一种新方法,也为临床用药研究提供了新方向.AIM: To use proteomics method find differential ex- pressed proteins of two-dimensional gel electrophoresis (2-DE) profiles of human hypertrophic scar, keloid and normal skin tissue, and to provide a new method for therapy of human pathological scars. METHODS: To establish and analyze two-dimensional gel electrophoresis (2-DE) profiles of hypertrophic scars, and normal skin. Identified part of differential expression proteins and identified by MALDI-TOF-MS. RESULTS: 2-DE profiles of human hypertrophic scar, keloid and normal skin tissue were compared 9 differential expression protein sports were selected and 5 differen- tial expression protein were identified; the proteins were: phos- phatidylethauolamine-binding protein, fatty acid binding protein, type I collagen, carboxy-propeptide of alpha Ⅲ procollagen, carboxy-propeptide of alpha I procollagen. CONCLUSION: In this study, 5 differential proteins among hmnan hypertrophic scar, keloid and normal skin tissue were identified. It may provide a new therapy and method system for study of the pathogenesis mechanism of pathological scars.
