摘  要:目的:观察生姜油对佐剂性关节炎(adjuvant arthritis,AA)小鼠血清细胞因子和T细胞亚群的影响。方法:将小鼠随机分成5组,即正常对照组、AA模型组、生姜油高剂量(1.0 ml/kg)组、生姜油低剂量(0.5 ml/kg)组、地塞米松(1.0 mg/kg)组。除正常对照组外,每只小鼠右后足跖皮下注射弗氏完全佐剂(FCA)制备AA小鼠模型。致炎后第8天起开始灌胃(ig)给药,1次/d,连续14d。分别在致炎后第8、12、16、20天测量小鼠足肿胀度,并于第21天处死小鼠,ELISA法测血清中IL-1β和IL-4,流式细胞仪测血液中CD4+和CD8+。结果:生姜油0.5、1ml/kg均能明显减轻小鼠足肿胀(P<0.05,P<0.01),生姜油1 ml/kg可显著降低AA小鼠血清IL-1β(P<0.05),但对IL-4无明显的影响,并阻止CD4+/CD8+升高。结论:生姜油对AA有抑制作用,其抗炎机制之一可能与降低血清IL-1β水平,抑制T细胞亚群CD4+/CD8+比例上升有关。Objective:To study Effects of ginger oil on serum cytokines and T lymphocyte in AA mice. Method:Mice were randomly divided into five groups:normal control, adjuvant arthritis (AA) model, hige-dose ginger oil(1.0ml/kg),low-dose ginger oil(0.5ml/kg)and dexamethasone. All mice were injected subcutaneously Freund’s complete adjuvant (FCA) in right voix pedis ,except for normal control. The intragastric administration procedure began at eight day after modeling and was once daily for fourteen days. The changes of paw swelling was measured by using volumetric method. Serum IL-1β and IL-4 was measured using ELISA. CD+4 and CD+8 was tested by flow cytometry. Result:Ginger oil could significantly inhibit paw swelling in AA mice at the dose of 1.0~0.5ml/kg. Ginger oil could remarkable diminish serum IL-1β at the dose of 1.0ml/kg,but had no significant effect on serum IL-4.Ginger oil could significantly prevent ratios of CD+4/ CD+8 from being rise which was induced by FCA. Conclusion:Ginger oil has inhibitory effect on AA . Diminishing serum IL-1β and balancing subgroup of T lymphocyte may be a part of the mechanism.
