摘 要:目的:探讨OPN反义基因(ANOPN)对人乳腺癌细胞系MDA-MB-231细胞生长及迁移能力的影响。方法:构建人骨桥蛋白反义基因真核表达质粒pcDNA3.1-ANOPN,将质粒pcDNA3.1-ANOPN、空载体pcDNA3.1(+)和等量的脂质体分别转染人乳腺癌细胞系MDA-MB-231,将转染细胞分别命名为MDA-ANOPN、MDA-vect和MDA,在体外观察各组细胞的迁移能力。结果:MDA-MB-231细胞比MDA-vect及MDA细胞迁移能力弱(P<0.01)。结论:ANOPN可抑制人乳腺癌细胞系MDA-MB-231细胞的迁移能力。Objective: To explore the effect of anti - sense gene of osteopontin (ANOPN) on migration ability of MDA - MB - 231 cells of breast cancer. Methods: Eukaryotic expression vector pcDNA3. 1 - ANOPN was constructed, then pcDNA3.1 - ANOPN, empty vector pcDNA3.1 ( + ) and commensurable liposome were used to transfect human breast cancer cell line MDA - MB - 321 cells, and the transfected cells were named as MDA - ANOPN, MDA - vect and MDA, respectively. The migration ability of these cells were observed. Results : The migration ability of MDA - MB - 231 cells was stronger than those of MDA - vest cells and MDA cells ( P 〈 0. 01 ) . Conclusion: ANOPN can suppress the migration ability of human breast cancer cell line MDA -MB -231 cells.
