摘  要:目的探讨整合素β1在肺癌发生、发展中的作用。方法应用基因芯片技术、基因表达的限制性分析(RAGE)法和流式细胞仪法检测整合素β1在肺腺癌细胞株A549、肺泡细胞癌细胞株H1650及小细胞肺癌细胞株DMS53中的表达情况。结果整合素β1在三种肺癌细胞中均出现高表达,以A549中表达最高,在非小细胞肺癌A549和H1650中表达显著高于在小细胞肺癌DMS53中的表达(P<0.05)。结论整合素β1在非小细胞肺癌侵袭及转移中可能发挥一定作用,有望成为肿瘤治疗的靶基因。Objective To investigate the role of integrin β1 in the occurrence and development of lung cancer. Methods The expression of integrin β1 in various types of lung cancer cells including A549 ( adenocarcinoma), H1650 ( bronchidoalveolar carcinoma), DMS53 (small-cell carcinoma) were detected by eDNA microarrays technique, restriction analysis of gene expression (RAGE) and flow cytometry. Results The expression of integrin β1 increased in the 3 types of lung cancer cells,among which the highest expression was found in A549 cell line, the expression in A.549 and H1650 cell line were significantly higher than that in the DMS.53 cell line( P 〈 0.05 ). Conclusions Integrin β1 may play certain effect in the intrusion and metastasis of non-small cell lung cancer, and it is likely to be a target gene for the tumor therapy.
