摘 要:目的探讨黄芪多糖对人乳腺癌MCF-7细胞株周期的影响及联用紫杉醇,对MCF-7细胞的杀伤作用。方法通过四甲基偶氮唑蓝(MTT)实验检测细胞的增殖抑制率,以观察黄芪多糖对MCF-7细胞的增殖抑制作用;应用流式细胞仪检测细胞周期是否有改变及凋亡率。结果黄芪多糖可抑制MCF-7细胞的生长、增殖;与紫杉醇联合应用对MCF-7细胞杀伤作用强于两种药物单独使用。黄芪多糖处理后的MCF-7细胞出现低于G1期DNA含量的亚二倍体凋亡峰,将细胞周期阻滞于G1期。结论黄芪多糖对MCF-7细胞有杀伤作用;黄芪多糖与紫杉醇联合使用具有协同抗肿瘤作用。Objective To study the effects of Astragalus Polysaccharides on MCF - 7 cell line cycle of mammary cancer and its killing action on MCF- 7 cell when in combination with paclitaxel. Methods A methylthiazolyltetrazolium (MTT) test was performed to measure the inhibition rate of cell proliferation so as to evaluate the inhibition of Astragalus Polysaccharides on MCF- 7 cell proliferation. A flow cytometer was used to test whether the cell cycles had changed or not and to measure the apoptosis rate. Results Astragalus Polysaccharides can inhibit the growth and proliferation of MCF- 7 cells. The killing action of Astragalus Polysaccharides in combination with paclitaxel on MCF- 7 cell is superior to Astragalus Polysaccharides alone or paclitaxel alone. After treatment with Astragalus Polysaccharides, hypodiploid apoptosis occurred which DNA content of MCF - 7 cells was lower than that in stage G1, the cells cycle was blocked in stage G1 Conclusion Astragalus Polysaccharides can kill MCF - 7 cells and its combination with paclitaxel can have synergic anti- neoplastic function.
