摘  要:目的:采用实时荧光定量PCR方法检测湿邪致病大鼠(TCRVβ)亚家族基因表达谱,从免疫识别的角度探讨湿邪致病"蒙蔽性"特点。方法:将32只SD大鼠按随机数字表随机分为正常对照组、内湿大鼠模型组、外湿大鼠模型组和内外湿大鼠模型组4组,造模20d后,各组按随机数字表选3只大鼠,检测其TCRVβ亚家族表达水平。结果:与正常对照组比较,外湿大鼠模型组的TCRVβ1、TCRVβ7、TCRVβ9、TCRVβ13表达升高(P<0.05,P<0.01,P<0.001)。与正常对照组比较,内湿大鼠模型组的TCRVβ8、TCRVβ14、TCRVβ18表达降低(P<0.05,P<0.01),内外湿大鼠模型组的TCRVβ8、TCRVβ18表达也降低(P<0.05,P<0.01)。结论:在外湿大鼠模型组部分TCRVβ亚家族比正常对照组表达升高,而在内湿大鼠模型组及内外湿大鼠模型组中部分TCRVβ比正常对照组表达低下,提示内湿之邪可能具有"蒙蔽性"有关,在内湿的环境中,机体可能对外在相关抗原相对不敏感,进而导致湿邪致病大鼠免疫功能障碍而死亡率升高,其机制值得进一步研究。Objective: To investigate variety pattern of expression level of TCRV β subfamilies in mononuclear cell in spleen tissue of rats with dampness pathogenic factors and normal rats by using FQ-PCR technique. Methods: 32 SD rats were divided into four groups: normal group, external dampness group, internal dampness group, external and internal dampness group. Observing period was 20 days. 3 Rats were randomly selected from each group in order to exam the TCRV β subfamilies expression level. Results: The expression of TCRV β 1, TCRV β 7, TCRV β 9 and TCRV β 13 in external dampness group were higher than those in normal group (P〈0.05, P〈0.01, P〈0.001). The expression of TCRV β 8, TCRV β 14 and TCRVβ 18 in internal dampness group were lower than those in normal group (P〈0.05, P〈0.01), also the expression of TCRV β 8 and TCRV β 18 in internal and external dampness group were lower than those in normal group (P〈0.05, P〈0,01). Conclusion: The expression of TCRV β subfamilies in dampness groups was disordered, several TCRV β expression in external group were higher while several TCRV β expression in internal dampness group and both internal and external group were lower than those in normal group, which mean the hoodwinks nature of dampness evil that might further cause immune disorder in model groups.
