摘  要:目的研究细菌内毒素定量法研究活性炭及超滤法的热原去除工艺。方法以动态浊度法测定不同浓度活性炭及不同截留分子量超滤膜使用前后药液中细菌内毒素的含量变化,观察其去除热原的效果。结果在三七总苷溶液中外加内毒素制备三七总苷原液,测得0.03%,0.10%,0.30%3种浓度活性炭的细菌内毒素去除率分别为62.21%,76.21%,81.03%;50000及100000相对截留分子量的超滤膜的细菌内毒素去除效率均>91%。结论该实验明确了不同浓度活性炭与其细菌内毒素去除效率的量化关系及超滤法去除细菌内毒素的效率,为注射剂生产工艺中去除热原提供依据。Objective To investigate the pyrogens removing technology with activated carbon and ultrafihration by quantitative detection of bacterial endotoxins. Methods The content of bacterial endotoxins was detected by keniticturbidimetry and the removal efficacy was studied before and after using different concentrations of active carbon and uhrafiltration membranes at different cut off molecular weight in medicine solution. Results The removal efficacy of bacterial endotoxins was 62.21%, 76.21%1 81.03% by using three concentrations of active carbon at 0.03% ,0. 10% ,0.30%, respectively in Panax Notoginseng Saponins solution, and it was above 91% with uhrafihration membrane at cut off molecular weight both 50 000 and 100 000. Conclusion The quantitative relationship between activated carbon and their bacterial endotoxins removal efficiency and which by uhrafiltration are clarified in our study. It provides evidences for removing pyrogens from injections in production technology.
