驱铅益智口服液对血液系统铅毒性的拮抗作用

摘  要:目的探讨驱铅益智口服液对血液系统安全性和铅染毒大鼠贫血的治疗作用。方法依据《食品安全性毒理学评价程序和方法》(GB 15193.1-2003)的要求,采用小鼠骨髓嗜多染红细胞微核实验评价驱铅益智口服液对血液系统安全性。以2%醋酸铅溶液灌胃3周,建立铅染毒大鼠模型,观察低、中、高剂量驱铅益智口服液(染毒后分别给于1 250、2 500和5 000 mg/kg体质量)治疗前后大鼠血铅含量、外周血血红蛋白浓度和血清铁蛋白的变化。结果低、中、高剂量驱铅益智口服液组雌雄小鼠的骨髓嗜多染红细胞微核率与阴性对照组相比差异均无统计学意义(P>0.05)。与模型对照组相比,3种剂量驱铅益智口服液均可显著下调铅染毒大鼠血铅水平(P<0.01),升高血红蛋白浓度(P<0.05或P<0.01),减低血清铁蛋白水平(P<0.01)。其效果优于或相当于乙二胺四乙酸的排铅作用。结论驱铅益智口服液对骨髓红系造血系统无明显致畸作用;并可显著减低血铅水平,上调铁储备,改善铅中毒大鼠贫血状态,拮抗铅对血液系统的毒性作用。Objective To explore the safty of Quqian Yizhi Health Oral Liquid(QYHOL) on blood system and the therapeutic effect on anaemia of lead-poisoned rats. Methods According to the requirements of The Toxicological Evaluation Procedure and Method of Food Safety(GB 15193.1-2003), the safety of QYHOL was studied by bone marrow polychromatic erythrocyte tiny nuclear test. Lead-poisoned rat models were established by garaging 2.0% lead acetate for three weeks. And the changes of blood lead concentration,haematoglobin and serum ferritin levels were investigated in lead-poisoned rats treated by low-, mid- and high-dose QYHOL. Results Data from bone marrow polychromatic erythrocyte tiny nuclear test showed that there were no differences between QYHOL groups and negative control group. Compared with model groups,three dosage QYHOL could decrease blood lead concentration (P〈0.01), increase haematoglobin concentration( P 〈0.05 or P 〈0.01) and down-regulate serum ferritin levels of lead-poisoned rats (P 〈 0.01). Conclusion QYHOL was a safe product without apparente toxicity on bone marrow erythropoiesis and could improve anaemia of lead-poisoned rats by expelling blood lead and increasing quantity of ferrum storage. This product could resist the toxicity of lead on blood system.
