摘  要:目的了解SD大鼠视网膜光损伤前后视网膜组织中一氧化氮(nitric oxide,NO)、超氧化物歧化酶(superoxide dismutase,SOD)及丙二醛(malondialdehyde,MDA)水平变化及其意义。方法 40只SD大鼠随机分成正常对照组、单纯光照组。单纯光照组SD大鼠应用乌拉坦麻醉后,使用缝线开睑法用非聚焦手术显微镜灯泡连续照射1h,调整光源与眼球表面距离使光照强度为(19 000.0±1 102.6)Lux,室内温度控制在25℃左右。光照后1、3、5、7 d分别取视网膜标本制备匀浆,分别进行视网膜光损伤后视网膜组织中的MDA、SOD及NO浓度测定。同时,正常对照组也取视网膜标本制备匀浆,进行视网膜组织中的MDA、SOD及NO浓度测定。结果光损伤后4个不同时间点正常对照组NO及MDA比单纯光照组降低,差异有统计学意义(P<0.01);正常对照组SOD比单纯光照组升高,差异有统计学意义(P<0.01)。结论①光照强度为(19 000.0±1 102.6)Lux的可见光能够引起大鼠视网膜光损伤;②通过应用抑制自由基产生和脂质过氧化药物可以起到保护视网膜光损伤的作用。Objective To observe the variation of nitric oxide(NO), superoxide dismutase (SOD) and malondialdehyde(MDA) in SD rat's retina before and after retinal photic injury. Methods Forty SD rats were randomly divided into two groups, control group and simple light-induced group(SL group). The rats of the SL group were anaesthetized with urethane,and their eyelids were opened by using suture. Then eyeballs were irradiated by non-focusing surgical microscope bulbs for an hour. The distance between the light and the eyeballs was adjusted in order to make the illumination intensity at (19 000.0±1 102.6) Lux. And the indoor temperature was regulated at 25℃. Afterwards,the retina tissue homogenate of the two groups were made and the level of NO, SOD and MDA, on 1,3,5,7 day after light-inducing, were detected. Menwhile, the retina tissue homogenate of the control group were made and the level of MDA, SOD and NO were detected. Results Independent sample t test were performed to compare NO,SOD and MDA of different groups respectively, and the result shows that NO and MDA of control group was lower than that of light-induced group, at four different time points, with significant difference ( P〈0.01); SOD of control group was higher than that of light-induced group, with significant difference ( P 〈0.01). Conclusion ①Visible light exposure of (19 000.0±1 102.6)Lux can lead to retinal photic injury in rats. ②The medicine which can inhibit free radicals generation and lipid peroxidation may protect retina from light damage.
