摘　　要：目的:采用"纤维蛋白原-凝血酶"法建立人胃癌组织块裸鼠胃癌原位移植模型,为研究胃癌的发病机制及实验治疗提供理想的动物模型。方法:以人胃癌细胞株SGC-7901反复接种传代于BALB/C裸鼠皮下形成的皮下移植瘤组织块为材料,采用"纤维蛋白原-凝血酶"法将其粘贴于裸鼠胃壁,观察原位移植瘤的生长情况,移植成功率及转移情况。结果:"纤维蛋白原-凝血酶"法人胃癌组织块原位移植成功率为100%,局部淋巴结转移率为100%,肝脏及其他脏器转移发生率为83%,腹腔积液形成率为50%。结论:"纤维蛋白原-凝血酶"法能成功地建立人胃癌组织块裸鼠原位移植模型,并能很好地重现胃癌患者的临床转移过程,可为胃癌的临床研究提供一种简便、易行的实验模型。Objective： To establish nude mouse orthotopic transplantation models of human stomach cancer constructed using intact tumor tissue by ＂Fibrinogen-thrombin＂ paste technique and to provide an ideal animal model for research in the pathogenesis and experimental treatment of stomach cancer. Methods： Transplanted gastric tumor tissue block was constructed through repeatedly subcutaneously inoculating and passaging human gastric carcinoma SGCo7901 cells in BALB/C nude mouse, and pasted orthotopically on the gastric wall of nude mouse with the ＂Fibrinogen-thrombin＂ paste technique. The orthotopic transplantation tumor growth characteristics , tumor -take rates and metastasis rates were examined. Results： A 100% successful tumor-take rate was obtained in the model and the metastasis rate of local lymph nodes , liver and other organ were 100% and 83% ,respectively. The rate of ascites formation was 50%. Conclusion： ＂Fibrinogen-thrombin＂ paste technique can establish nude mouse orthotopic transplantation models of human stomach cancer constructed using intact tumor tissue successfully and will represent clinical human stomach cancer metastasis process excellently. It can provide an simple and ideal animal model for clinical studies of stomach cancer.
