摘  要:目的探讨体外情况下IL-1β对血管平滑肌细胞组织蛋白酶S(Cathepsin S,Cat-S)的表达影响。方法原代培养小鼠主动脉平滑肌细胞(VSMC)传代后用IL-1β(10ng/ml)诱导细胞12、24、36h,免疫荧光(Immunofluorescence,IMF)法检测VSMC Cat-S的表达变化,ELISA试剂盒检测细胞培养基Cat-S浓度。结果原代培养的细胞VSMC-α-actin呈阳性,细胞纯度达96%以上。Cat-S免疫荧光染色,对照组呈阴性,IL-1β组均为阳性,并随着诱导时间的延长荧光强度不断增强。对照组培养基Cat-S浓度微量,经IL-1β诱导12 h后明显增加,与对照组相比差异显著(P<0.01),24 h后达到高峰并趋于稳定。结论炎性细胞因子IL-1β在体外可促进VSMC表达、分泌组织蛋白酶Cat-S。Objective To study the influence of IL-1β on expression of Cathepsin S in VSMC in vitro. Methods Mouse VSMC was cultured by primary methods. After passage culture, to incubate VSMC with IL-1β (10ng/ml) for 12h, 24h, 36h; then detect the Cat-S expression in VSMC by IMF and test the concentration of Cat - S in medium by ELLISA methods. Results IMF showed that primary culture cells have positive VSMC - actin staining and the cell purity was more than 96%. Cat - S showed negative immunofluoreseence staining in control VSMC but positive staining in VSMC induced with IL-1β and the staining intensity grown up following the induced time last. The Cat - S concentration in medium increased clearly after VSMC induced by IL - 18 for 12h and reached the peak after 24h then keeping this level. The change has significant statistic difference( P 〈 0.01 ). Conclusion IL-1β can induce VSMC to express Cat- S in vitro.
