癌-睾丸抗原SSX基因家族在视网膜母细胞瘤中的表达研究

摘　　要：目的检测癌-睾丸抗原SSX1-5 mRNA在视网膜母细胞瘤（RB）组织中表达情况,以及用于RB免疫治疗的可能性。方法采用逆转录-聚合酶链反应（RT-PCR）技术对18例RB组织、18例非肿瘤病变视网膜组织及18例眼部正常组织中SSX1-5 mRNA的表达进行分析。结果18例RB组织中,SSX1、SSX2和SSX4 mRNA的表达率分别为33.3%（6/18）、5.6%（1/18）和22.2%（4/18）;未检测到SSX3和SSX5的表达。RB组织中,至少表达1个SSX基因者为50.0%（9/18）;表达2个或2个以上者为11.1%（2/18）。在非肿瘤病变视网膜组织以及眼部正常组织中无1例表达SSX1-5 mRNA。SSX1-5基因的表达与年龄、性别、肿瘤大小、分化程度和临床分期等均无显著相关性（P〉0.05）。结论SSX1-5在RB组织中表达频率不尽相同,但具有较强的特异性,提示该基因家族有可能成为RB免疫治疗的靶抗原。Objective To investigate the expression of cancer-testis antigens（CTA） SSX1-5 mRNA in retinoblastoma（RB） and the possibility of applying these antigens as targets for specific immunotherapy for RB.Methods Expression levels of CTA SSX1-5 mRNA were detected with reverse transcription polymerase chain reaction（RT-PCR） in 18 RB tissues,18 non-tumor retinal tissues and 18 normal eye tissues.Correlation between the expression of CTA SSX1-5 and clinical data was evaluated.Results Of 18 RB tissues,SSX1,SSX2 and SSX4 mRNA were expressed in 33.3%（6/18）,5.6%（1/18） and 22.2%（4/18）,respectively.About 50.0%（9/18） and 11.1%（2/18） of RB tissues positively expressed at least one and two members of SSX1-5,respectively.In addition,no samples of non-tumor retinal tissues and normal eye tissues expressed SSX1-5.No relationship was found between the expression of CTA and clinical indicators such as age,gender,tumor size,differentiation and clinical stage（P〉0.05）.Conclusion CTA SSX1-5 are expressed with different percentage in RB,but with high specificity,which are possibly promising targets for antigen-specific immunotherapy of RB.
