摘 要:目的观察大鼠实验性脑挫伤后不同时间内脑组织核转录因子(NF-κB)表达的变化,探讨NF-κB与脑损伤经过时间的关系。方法参照Feeney’s法建立大鼠脑挫伤模型,在伤后1h,4h,12h,48h,72h,7d,14d,运用Western blot法分别检测NF-κB的表达量,以非损伤组做为对照。结果NF-κB表达在损伤后1h出现,12h即达到高峰,48h开始下降,7d后表达量仍高于对照组,14d后表达量基本恢复至接近对照组水平,各实验组与对照组比较差异有统计学意义(P<0.05)。结论NF-κB在脑挫伤后的表达经历了一个先快速升高后又下降到接近正常水平的过程,其表达的时序性变化可望成为临床法医学脑挫伤的早期鉴定及推断脑损伤时间的指标之一。Objective To investigate the changes of NF-κB expression in different time after the experimental traumatic brain injury in rats,and to explore its relationship with the brain damage time. Methods Based on Feeney' s method,the rat model of cerebral contusion was established. At 1 h,4 h, 12 h,48 h,72 h ,7 d, 14 d after brain injury, the expression of NF-κB was detected by Western blot. Eight rats without brain injury were chosen as controls. Results The expression of NF-κB was found at 1 h after injury, increased at 4 h,peaked at 12 h,and returned to the level of controls at 14 d. Little expression was found at 7 d,but still higher than that of the controls. Conclusion Sequential changes of NF-κB expression after cerebral contusion suggest that the expression of NF-κB may be served as a marker for early diagnosis and timing of cerebral contusion in forensic practice.
