摘  要:目的:探讨血管性痴呆小鼠海马神经元JNK的表达及其意义。方法:应用双侧颈总动脉线结反复缺血-再灌注法制备血管性痴呆小鼠模型,并设正常组、假手术组、血管性痴呆模型组(模型组),利用跳台试验和水迷宫试验观测其行为学改变,应用免疫组化技术观测其海马神经元JNK的表达变化。结果:模型组小鼠与正常组、假手术组相比较,模型组小鼠学习成绩为跳台试验反应时间长,游全程时间长,错误次数多。记忆成绩为跳台试验潜伏时间短,游全程时间长,错误次数多(P<0.05)。海马的JNK免疫反应阳性细胞元免疫组化评分明显增加(P<0.01)。结论:血管性痴呆小鼠认知功能、记忆功能减退与JNK表达增加有关。Objective To explore the expression and the significance of JNK in neuron of hippoeampus in mice with vascular dementia(VD) and its role in the development of VD. Methods 66 mice were divided into three groups (the normal group, sham operation group, and the VD models group). The mice of VD models group were subjected to isehemia and reperfusion repeatedly by ligation of bilateral common carotid arteries to establish the VD models. The behavioral changes of the mice were investigated by step-down test and morris water maze test. The Changes of immunohistochemical expression of JNK were observed in the three groups. Results Compared with the normal group and sham operation group, the learning test showed that the response time and the swimming time of VD models group were prolonged, and the error times were also increased; In the memory test, the latency time of VD models group was shortened, the swimming time was prolonged, and the error times were increased (P 〈 0.05). The immunohistochemical expression of JNK in neuron of hippoeampus in VD models group was also increased, compared with the other two groups (P 〈 0.01 ). Conclusion It suggests that the decline of cognition and memory function is related to the increasing of JNK expression in neuron of hippoeampus in VD mice.
