摘 要:目的:探究异氟烷对幼龄小鼠生长和学习记忆的影响。方法:按分层随机分组将64只幼龄小鼠分为两大组,每大组各分为4小组(n=8),即NS组(生理盐水组)、Iso1组、Iso2组和Iso3组(后3组异氟烷剂量分别为0.05、0.1、0.2mL/kg).用跳台实验和避暗实验分别测定给药后1、2、3、5d的错误次数和潜伏期以及注药前基础体重和注药后1d至6d各组幼龄小鼠体重。结果:在跳台实验中,与NS组相比,Iso1组和Iso2组在1、2、3、5d错误次数略增;Iso1组、Iso2组和Iso3组潜伏期在1、3、5d缩短,尤其在1d,Iso2组和Iso3组潜伏期明显缩短(P〈0.05或P〈0.01);各组幼龄小鼠在1~6d体重大致呈增长趋势(P〈0.05或P〈0.01),与NS组相比,Iso1组、Iso2组和Iso3组在1—4d体重变化量呈降低趋势,尤其在1d有明显降低(P〈0.01)。在避暗实验中,与NS组相比,Iso1组、Iso2组和Iso3组错误次数在1—3d略增;Iso1组在1、2、3、5d潜伏期略缩短,Iso2组在3d潜伏期略缩短,Iso3组在1、2、3d潜伏期略缩短。与NS组相比,Iso2组和Iso3组在1~4d幼龄小鼠体重增长速度在2~3d降低(P〈0.05或P〈0.01)。结论:异氟烷可在短期内损害幼龄小鼠学习记忆能力和生长,对长期学习记忆能力和生长均无明显影响。Objective: To investigate the effect of isoflurane on learning , memory and growth in young mice. Methods:The young mice were randomly divided into two large groups,each large group were rangdomly divided into four groups (n =8) :normal saline group(NS group), Iso1 group,Iso2 group and Iso3 group(the doses of last three are respective 0. 05,0. 1,0. 2 mL · kg^- 1 ). The number of error and the length of latency were recorded one day, two days, three days and five days after the drug administration in the step - down and step - through tasks and the weights of young mice were recorded from 1 st to 6th day after the drug administration. Results: In the step - dawn tasks, as compared with the NS group, on the first day, second day, third day and fifth day , the number of error was increased in the Isoj and Iso2 groups. On the first day, third day and fifth day,the length of latency decreased in the Iso1 , Iso2 and Iso3 groups,especially on the first day, the length of latency decreased obviously in the Iso2 and Iso3 groups( P 〈 0. 05 or P 〈 0. O1 ) ;The weigh of all groups showed the tendency of increase on the first day to the sixth day(P 〈 0. 05 or P 〈 0. 01 ). As compared with the NS group, on the first day to forth day,the number of the changes of the weighs showed a tendency of decrease, especially on the first day( P 〈 0. 01 ). In the step - through tasks, as compared with the NS group, on the first day to third day, the number of error was increased in the Iso1, Iso2 and Iso3 groups, on the first day, second day,third day and fifth day,the length of latency was decreased in the ISOl group, the length of latency of Iso2 group decreased on the third day ;the length of latency of Iso3 group decreased on the first day to third day. As compared with the NS group, on the first day to forth day, the gvelocity of the growth decreased on the second day to the third day( P 〈 0. 05 or P 〈 0. 01 ).
