摘  要:目的观察黄芪甲苷、β-榄香烯对人胃癌细胞中COX-2、VEGF和PGE2表达的影响。方法采用MTT试验观察黄芪甲苷、β-榄香烯对胃癌细胞的抑制作用,用RT-PCR以及Western blot方法研究加药后人胃癌细胞C0X-2基因及其蛋白表达的变化,用ELISA方法检测培养基中VEGF、PGE2的表达情况。结果黄芪甲苷、β-榄香烯能抑制人胃癌细胞生长,呈一定的量效特征;并能抑制人胃癌细胞COX-2、VEGF和PGE2的表达。结论黄芪甲苷、β-榄香烯抗肿瘤的机制可能是通过抑制COX-2,进而抑制其下游产物PGE2的表达及使VEGF表达下调,从而抑制肿瘤的生长。Objective It is to observe the effect of Astragaloside and β - elemene on the expression of COX - 2, vascular endothelial growth factor(VEGF) and prostaglandin E2 (PGE2 ) in human gastric cancer cell. Methods The inhibitory effect of Astragaloside and β- elemene on viability of SGC7901 cell was assessed by MTT assay. The expression of COX 7 2 mRNA and protein was determined by RT- PCR and Western blot respectively. The expression of VEGF and PGE2 in culture medium was detected by ELISA. Results Astragaloside and β- elemene inhibited growth of SGC7901 cell in a dose-dependent manner and down-regulated expression of COX - 2,VEGF and PGE2. Conclusion The anti-tumor mechanism of Astragaloside and β - elemene is related with inhibition of COX - 2 expression, which causes the suppression of its downstream product PGF2 expression and down-regulation of VEGF, therefore decreases tumor growth.
