摘  要:目的以鼻咽癌细胞株CNE为靶细胞,探讨CIK细胞对鼻咽癌细胞的杀伤作用。方法正常人外周血单个核细胞在体外经CD3单抗、rhIFN-γ、rhIL-1多种细胞因子诱导后所收获的CIK细胞作为效应细胞,以鼻咽癌细胞株CNE1和CNE2为靶细胞。用MTT法测定效应细胞对靶细胞的杀伤活性。结果CIK细胞经体外诱导培养12 d后,CD3+细胞、CD8+CD28+细胞和CD3+CD56+细胞的阳性百分率分别为(92.69±5.69)%、(38.01±14.63)%和(27.74±7.96)%。在效靶比为30%时,CNE1和CNE2的杀伤活性分别为(67.90±8.19)%和(65.20±3.80)%,两者比较差异无统计学意义(P>0.05)。结论CIK细胞可用于鼻咽癌放射治疗后的补充治疗。该细胞对CNE1和CNE2两种不同分化程度的鼻咽癌细胞具有相似的杀伤活性,提示临床上利用CIK细胞进行补充治疗,无须考虑细胞的分化程度。Objective To investigate the killing activity of cytokine induced killer cells against nasopharyngeal carcinoma cells CNE as the target cells. Methods PBMCs were isolated from healthy human venous blood, then were induced to be CIK cells in vitro as the effect cell by the presence of anti-CD3 antibody ,rhIFN-3, and rhlL-1. With CNE1 cells or CNE2 cells as the target cell. Then the killing activity of effect ceils against target cells was measured by MTT method. Results The CIK cells were analyzed by flow cytometry in vitro after 12 days. The positive percentage ofCD3^+, CD8^+CD28^+, CO3^+CD56^+ cell were (92.69±5.69)%, (38.01±14.63)%, (27.74±7.96)% respectively. At the effect target ratio 30% , the killing activity of CIK cells against CNE1 cells (67.90±8. 19) % was not significantly higher than that of CNE2 cells (65.20±3.80) % ( P 〉 0.05 ). Conclusion The research has provided the basis for the immunotherapy of CIK cells against nasopharyngeal carcinoma after radiotherapy. The killing activity of CIK cells against CNE1 cells is the same as that of CNE2 cells. So, we need not consider the differentiation degree of cells with CIK cells in clinic.
