摘　　要：目的探讨基质金属蛋白酶-2(MMP-2)及金属蛋白酶组织抑制剂(TIMP-2)在人非小细胞肺癌(NSCLC)中的表达及其与NSCLC转移、预后的相关性,分析NSCLC发生、发展、侵袭和转移的机制。方法对MMP-2和TIMP-2的蛋白进行间接免疫荧光标记,采用流式细胞仪对60例非小细胞肺癌及20例癌旁正常肺组织进行定量检测。结果MMP-2、TIMP-2蛋白及MMP-2/TIMP-2在非小细胞肺癌组织中的表达量明显高于癌旁正常肺组织(P<0.05);MMP-2蛋白表达量和MMP-2/TIMP-2比值与肺癌分化程度、TNM分期、淋巴结转移及术后生存期有关(P<0.05);TIMP-2蛋白表达与患者临床病理特征均无关(P>0.05)。结论在肺癌发生中,MMP-2表达量上调,MMP-2/TIMP-2的失平衡,MMP-2蛋白过剩,是导致非小细胞肺癌发生的主要原因。可作为非小细胞肺癌恶性程度及预后判断的指标。Objective To investigate the expression of matrix metalloproteinase 2 （MMP-2）and tissue inhibitor of metalloproteinase 2 （ TIMP-2 ）, and to explore their correlation with metastasis and prognosis in non-small-cell lung carcinoma. Methods The expression of MMP-2 and TIMP-2 in 60 cases of lung cancer tissues and 20 cases of paracancerous tissues of lung was examined by flow cytometry. Results The levels of MMP-2, TIMP-2 and MMP-2/TIMP-2 were significantly higher in lung cancer tissues than those in paracancerous normal lung cancer tissues （ P 〈 0.05）. The expression of MMP-2 and the ratio of MMP-2/TIMP-2 were significantly correlated with the differentiation,TNM stage,lymphatic metastasis and patients＇ survival time after operation （ P 〈 0.05）, however, the expression of TIMP-2 was not correlated with the clinicopathological characteristics of patients with lung cancer（ P 〉 0.05）. Conclnsion The overexpression of MMP-2, the imbalance of MMP-2 and TIMP-2 are major reason for resulting in the cancer pathogenesis, which can be used as the reference indexes for the malignant degree of non-small-cell lung carcinoma and patients＇ prognosis.
