摘 要:目的利用高脂饲料和四氯化碳(CCl4)复合诱导小鼠非酒精性脂肪肝(NAFLD)模型。方法40只昆明小鼠随机均分为4组:普通饲料+生理盐水(A)组、高脂饲料+生理盐水(B)组、普通饲料+CCl4稀释液(C)组、高脂饲料+CCl4稀释液(D)组,3周后,比较各组血清生化指标及肝组织病理学指标的差异。结果D组小鼠肝指数[(5.5±0.18)%]、TG(2.01±0.27)mmol/L、TC(5.35±0.73)mmol/L、ALT(389.28±18.02)U/L、AST(416.86±30.68)U/L,均明显高于A、B、C组(P<0.05);C组小鼠血清ALT、AST明显高于A组(P<0.05)。镜下D组大部分肝细胞脂肪变性,脂滴大量成群出现且严重聚积,与其他各组差异显著。结论通过3周的高脂饲料与CCl4复合诱导,可快速建立小鼠NAFLD模型。Objective To establish a non-alcoholic fatty liver model(NAFLM)in mice by high-fat feeding and carbon tetrachloride.Methods Fourty Kunming mice were randomly divided into 4 groups with 10 mice each.The mice were fed with common food plus normal saline(group A),high-fat plus normal saline(group B),common food plus carbon tetrachloride(group C),or high-fat food plus carbon tetrachloride(group D).After three weeks,all mice were killed.The liver index,serum biochemical criteria and hepatic histological changes were compared.Results The liver index [(5.5±0.18)%)],serum TG [(2.01±0.27)mmol/L],TC [(5.35±0.73(mmol/L],ALT [(389.28±18.02(U/L],AST [(416.86±30.68(U/L] of group D were all higher than those of groups of A,B,C(P〈0.05).Compared with group A,the serum AST and ALT of group C were higher those of group A(P〈0.05).Compared with other groups,a lot of hepatocellular diffuse steatosis and a large quantity of the fat drop appeared and accumulated seriously in group D.Conclusion The NAFLM of mice can be established successfully by feeding with high-fat food and injecting carbon tetrachloride for 3 weeks.
