摘  要:探讨以白细胞介素2(IL-2)基因修饰的肿瘤细胞作为瘤苗治疗膀胱癌的应用价值。采用脂质体介导法将携带IL-2基因的逆转录病毒载体转染入膀胱癌细胞系BTT739中,流式细胞仪行细胞DNA周期分析。动物实验观察放射线灭活后基因瘤苗细胞的抗肿瘤免疫作用。建立能分泌IL-2的膀胱癌瘤苗BTT739/IL-2,DNA周期分析表明IL-2基因的导入及表达对BTT739细胞的增殖无影响。放射线灭活后BTT739/IL-2细胞丧失增殖能力,但仍能维持一定水平的IL-2分泌活性达2周左右。先用灭活的基因瘤苗免疫小鼠,可诱导免疫保护,3周后接种BTT739不形成肿瘤;用灭活的基因瘤苗治疗荷瘤小鼠,可使50%小鼠肿瘤消退并长期存活;对治愈后长期存活的小鼠再次接种高剂量BTT739细胞,仍无肿瘤形成。说明转染IL-2基因的膀胱癌瘤苗诱导的抗肿瘤作用对预防肿瘤的发生、抑制和清除微小瘤灶,防止膀胱癌复发具有重要的应用价值。To explore the value of IL 2 gene modified tumor cells as a vaccine for the treatment of bladder cancer, the retroviral vector carrying IL 2 genes was transfected into murine bladder cancer cell line BTT739 by Lipofectamine method. Cell DNA cycle was analysed by FCM and anti tumor immunity induced by inactivated gene vaccine was observed in experimental models. The results showed that the auto secretin IL 2 bladder cancer vaccine(BTT739/IL 2) was established.FCM analysis of cell DNA contents indicated that the IL 2 gene transfer and expression would not influence the proliferation of the gene modified cells.With X ray irradiation in vitro , growth of the cells was abrogated but its ability to secrete IL 2 was retained within 2 weeks. The anti tumor effect of the IL 2 gene transferred bladder cancer vaccine may have an important therapeutic value in the treatment of bladder cancer.
