摘  要:目的:观察聚乙烯醇(polyvinyl alcohol,PVA)对大鼠牙髓细胞的矿化诱导能力。方法:制备PVA涂层24孔板,分别以PVA、矿化诱导液对第4代SD大鼠牙髓细胞进行矿化诱导,通过定量的茜素红染色分析和碱性磷酸酶(ALP)活性检测比较二者的矿化诱导能力。结果:经PVA诱导的大鼠牙髓细胞,其ALP活性和茜素红分析在7 d的培养过程中,显示明显持续增高的趋势(P<0.01)。结论:PVA可诱导大鼠牙髓细胞矿化分化,有望成为组织工程化人工牙研究的支架材料。AIM: To investigate the mineralization inducing effect of polyvinyl alcohol (PVA) on dental pulp cells of SD rat . METHODS: Primary dental pulp cells of rat were cultured on non-coated (control), non-coated plus mineralization solution and PVD-coated 24 well culture plates in DMEM. The mineralization of the cells was determined by measuring the alkaline phosphatase (ALP) activity and by Alizarin Red S (ARS) mineralization assay at 1,4 and 7d after induction. RESULTS: At all time points, both PVA and mineralization inducing solution significantly increased ALP activity and calcium sedimentation ( P 〈 0.01 ) of the cells. PVA showed much stronger differentiation inducing effect than the mineralization-inducing solution (P 〈 0.01 ). CONCLUSION: PVA is able to induce the differentiation and mineralization of rat dental pulp cells and might be a suitable scaffold for the study of tissue engineering teeth.
