摘  要:目的:研究IFN-γ对卵巢癌耐药细胞株SKOV3TS及敏感细胞株SKOV3中STAT-1表达的调控。方法:应用RT-PCR方法、细胞免疫荧光法检测SKOV3TS与SKOV3在IFN-γ作用前后STAT-1 mRNA及蛋白的表达。用MTT法检测不同浓度IFN-γ作用卵巢癌细胞株不同时间后对细胞增殖率的影响。结果:卵巢癌SKOV3TS、SKOV3细胞株均有STAT-1表达,且主要集中于胞浆。并且随着IFN-γ作用时间延长,STAT-1蛋白表达增强。IFN-γ作用两种细胞株24h及48h后STAT-1 mRNA表达均增加,且差异有统计学意义(P<0.01),但不同浓度IFN-γ作用相同时间后STAT-1 mRNA表达差异无统计学意义(P>0.05)。IFN-γ可显著抑制SKOV3TS和SKOV3细胞增殖,也呈时间依赖性(P<0.01)。IFN-γ对SKOV3组的抑制率较SKOV3TS组高(P<0.05)。结论:IFN-γ能够促进STAT-1表达,抑制卵巢癌细胞增殖。Objective:To study the effect of recombinant human IFN-γ on the expression of STAT-1 in ovarian cancer cell lines.Methods:It is designed to detect the expression of STAT-1 in mRNA and protein levels in SKOV3 and taxol-resistant ovarian cancer cell line SKOV3TS after interference of recombinant human IFN-γ using RT-PCR and cell immunofluorescence.MTT was used to examine the influence of IFN-γ with different time duration and different concentration to the proliferation of ovarian caner cell lines.Results:Expression of STAT-1 protein was observed in the cytoplasm of SKOV3TS and SKOV3.And with the increase of time duration of IFN-γ,protein expression of STAT-1 in both of cell lines raises.When the cell lines cultured under the IFN-γ in 24 hours and 48 hours,the mRNA expression of STAT-1 was significantly improved(P〈0.01).But under the different concentration of IFN-γ,the mRNA expression of STAT-1 was not significantly changed(P〉0.05).IFN-γ can inhibit SKOV3TS and SKOV3 cell growth and proliferation,which showed the trend of time-dependent(P〈0.01).The inhibits rate of SKOV3 was higher than that of SKOV3TS.Conclusion:IFN-γ can enhance STAT-1 expression.It has remarkable inhibition effect on proliferation of ovarian cancer cells.
