人肝癌组织中原癌基因Pokemon的表达及其临床意义

摘　　要：目的探讨肝癌组织中Pokemon基因的表达,并分析其与肿瘤的大小、分化程度、TNM分期等临床资料的相关性。方法收集30例原发性肝癌患者的肿瘤及其癌旁组织,用逆转录酶聚合链反应(RT-PCR)及免疫印迹(Western blot)的方法分别从核酸及蛋白水平检测标本中Pokemon基因的表达,并对其与患者的性别、肿瘤大小、淋巴结转移、肿瘤分化程度、肿瘤的TNM分期等临床资料进行统计分析。结果RT-PCR检测肝癌和癌旁组织mRNA表达的相对强度分别为0.71±0.03、0.17±0.01。Western blot检测蛋白水平的相对表达强度分别为10.64±0.92、1.06±0.18,两者比较有统计学意义(P<0.05),肝癌组织中Pokemon的表达与肿瘤的大小有关,而与患者的年龄、肿瘤的分化程度、淋巴结转移及肿瘤TNM分期等无关。结论Pokemon的表达上调在肝癌的增殖中可能发挥重要作用。Objective To explore the expression of Pokemon in human hepatocellular carcinoma and its correlation with clinical features.Methods RT-PCR and Western blot were applied to detect the expression of Pokemon in 30 cases of resection samples and their corresponding paracancerous samples.Correlation between expression of Pokemon and clinical features was analyzed.Results The expressions of Pokemon mRNA and protein were significantly higher in the cancerous tissues than those in the paracancerous tissues （P〈0.05）.The expression of Pokemon was closely related to the size of samples,but no correlation with patient＇s sex,tumor differentiation degree,lymphoid metastasis and TNM stages.Conclusion Pokemon is expressed in human hepatocellular carcinoma and it may play a key role in the proliferation of cancer.
