摘 要:目的探讨血管内皮生长因子(VEGF)-A_(165)及血管生成在鼻内翻性乳头状瘤(NIP)癌变中的临床意义,评价VEGF-A_(165)与CD105标记的微血管面数密度(MVNA)的相关性。方法检测CD105免疫荧光标记的正常对照组(9例)、NIP组(23例)及NIP癌变组(8例)的MVNA,应用Western印迹法检测VEGF-A_(165)的表达,采用Spearman相关系数对VEGF-A_(165)蛋白表达量与CD105-MVNA的相关性进行分析。结果正常对照组、NIP组、NIP癌变组的MVNA依次增高,分别为(33.91±17.88)、(123.35±26.02)、(171.86±49.37)个/mm^2,3组间差异均有统计学意义(P值均<0.01);VEGF-A_(165)蛋白表达依次增高,分别为(94 927.9±8 633.5)、(110 653.2±8 419.7)、(11 6357.1±3 926.5)灰度值/mm^2,3组间的差异均有统计学意义(P值均<0.05)。VEGF-A_(165)与CD105标记的MVNA呈明显正相关(r=0.735,P<0.05)。结论血管生成在NIP癌变中有重要的作用,VEGF-A_(165)与CD105标记的肿瘤MVNA呈正相关,是NIP癌变的预示指标,能为NIP的临床治疗提供参考。Objective To assess the clinical significance of vascular endothelial growth factor (VEGF)- A165, angiogenesis in malignant transformation of nasal inverted papilloma(NIP), and to evaluate the correlation between VEGF-A165 and microvessel number of per unit area (MVNA) labeled by CD105. Methods Frozen sections of patients with normal mucosa (9 cases), NIP (23 cases), and squmamous cell carcinoma (SCC) derived from NIP (8 cases) were studied. Serial sections were stained with CD105 by immunofluorescence, and MVNA values were counted. Western blot analysis was used to measure the expression of VEGF-A165 The correlation between VEGF-A^6sand MVNA value was studied. Results The MVNA values was 33.91 ± 17.38 in normal mucosa group, 123.35 ±26.02 in NIP group, and 171.86± 49.37 in SCC group, and there were significant differences between the three groups (P〈0.01). VEGF-A165 expression densities (absorbance per mm^2 ) in the normal, NIP, and SCC groups were 94 927.9±8 633. 5, 110 653.2±8 419.7,and 116 357. 1±3 926.5, respectively, with significant differences found between the three groups (P〈0.05). VEGF-A165 was positively correlated with CD105-labelled MVNA, (r = 0. 735, P〈0.05). Conclusion Angiogenesis plays an important role in NIP malignant transformation. VEGF-A165 is positively correlated with CD105-Labelled MVNA value. VEGF-A165 and CD105-labelled MVNA value may serve as important indicators to evaluate NIP malignant transformation.
