摘 要:目的探讨叶酸脂质体对乳腺癌4T1细胞的体内靶向性,为开展叶酸介导的靶向性抗肿瘤药物的研究奠定基础。方法采用薄膜分散法结合冷冻超声法制备叶酸脂质体,原子力显微镜(AFM)确定叶酸脂质体的粒径大小和表征叶酸脂质体的表面形态结构。采用鼠源4T1细胞建立小鼠原位乳腺癌模型,原位皮下分别注射包封钙黄绿素的叶酸脂质体和脂质体,用荧光显微镜定性观察和荧光定量法测定叶酸脂质体与脂质体对肿瘤细胞的富集量。结果叶酸脂质体富集于原发性和转移性乳腺癌细胞的数量明显高于脂质体(P<0.05)。结论通过细胞表面叶酸受体的介导作用,叶酸脂质体能明显富集于表达叶酸受体的乳腺癌细胞上,即具有肿瘤靶向性;本研究为开展叶酸介导的靶向性抗肿瘤药物的研究提供了依据。Objective To investigate the targeting effect of folate liposomes to 4T1 breast cancer cells in vivo, and to lay the foundation for future study of folate-mediated anti-tumor drugs. Methods Folate hposomes were prepared by thinfilm dispersion method and frozen ultrasound method, then the particle size and surface morphological structure of which were determined by atomic force microscope (AFM). Breast cancer mice established with murine 4T1 cells were given wrapped Calcein folate liposomes and liposomes by subcutaneous injection around tumor. The diversity of the concentration between folate liposomes and liposomes in tumor cells were observed by fluorescence microscopy. Results The amounts of folate liposomes enriched in primary and metastatic breast cancer cells were significantly greater than that of liposomes ( P 〈 0.05 ). Conclusions Through the role of folate receptor mediated in the cell surface, folate liposomes can be significantly enriched in breast cancer cells expressing folate receptor highly. This study provides the basis for the further research of folate-mediated anti-tumor drugs.
