摘 要:目的检测重型β地贫患儿外周血miR-144基因表达水平,并分析miR-144与珠蛋白比率变化的关系。方法运用RT-PCR分别检测各20例重型β地贫患儿与健康对照组外周血miR-144基因表达水平及α/β珠蛋白的比率变化。结果与健康对照组比较,重型β地贫患儿外周血miR-144表达水平下降,差别有统计学意义,并且与α/β珠蛋白基因比率变化呈负相关性。结论微小RNAmiR-144可能调控重型β地贫患儿α/β珠蛋白基因比率的变化,将为重型β地贫的基因调节治疗提供新的实验依据。Objective To detect the expression levels of miR-144 gene in peripheral blood of children with β- thalassemia major and analyse the relationships between miR-144 gene and the variation of globin . Methods The level of miR-144 in peripheral blood and variation of α/β-globin gene in 20 children with β-thalassemia major and 20 healthy controls were detected by RT-PCR. Results Compared with the healthy controls,the levels of miR-144 in in peripheral blood of children with β-thalassemia major decreased,showing statistically significant difference and the level of miR-144 was negatively correlated with the cariations of α/β-globin gene. Conclusion miR-144 may involes the regulation of changes of α/βglobin gene in children with β-thalassemia major,which may provide a new experimental basis for gene treatment of β-thalassemia major.
