摘  要:以香石竹花蕾的子房壁为材料,研究了不同大小花蕾和预处理方法对香石竹染色体制片效果的影响。结果表明:幼嫩花蕾的子房壁中期分裂相细胞多;在3种预处理方法中,用冰水混合物预处理24 h效果最好;与根尖相比,幼嫩子房壁中处于分裂中期的细胞较多,且中期染色体在形态上与根尖中的无明显区别;利用子房壁制片,验证了13个香石竹品种的染色体数目,多数为30(二倍体)。The ovary walls of carnation buds were used to study the effects of different sizes of buds and pretreatment methods on the slide - preparation of the chromosomes in carnation. The results revealed that the ovary wall of young buds had more metaphase cells ; the pretreatment with mixture of ice and water for 24 hours had the best result among three methods of pretreatment. In compari- son with the root tip, the immature ovary wall possessed more cells which were at metaphase stage. There was no obvious difference in the morphology of metaphase chromosomes in both materials. Through chromosome preparations by using ovary wall, the chromosome number of 13 carnation cultivars was confirmed, the results showed that the chromosome number of most carnation cultivars was 30 (diplont).
