膀胱肿瘤中EphA2 mRNA的表达及其生物学行为关系研究

摘　　要：目的研究受体酪氨酸激酶EphA2在膀胱肿瘤中的mRNA表达,并对其致癌机制加以初步探讨。方法应用实时荧光定量RT-PCR技术检测49例膀胱尿路上皮癌(bladder urothelial carcinoma,BUC)、19例膀胱内翻性乳头状瘤(inverted papilloma of blader,IPB)和10例正常膀胱组织标本中EphA2 mRNA的表达,并通过2-△△CT方法分析相对基因表达的差异。结果相对于正常膀胱组织,EphA2 mRNA在IPB和BUC中的表达量明显增加,分别平均上调约2.06(2.06±0.84)倍和2.57(2.57±0.97)倍;在IPB与BUC组间的表达差异无统计学意义(P>0.05)。EphA2 mRNA的表达与膀胱肿瘤临床分期(r=0.471,P=0.001)及病理分级(r=0.286,P=0.047)呈正相关,与BUC其它各临床病理特征之间没有相关性(P>0.05)。结论EphA2过表达可能促成了IPB的癌性转化以及膀胱肿瘤的恶性进展,其核酸调控机制可能发生在膀胱肿瘤早期,甚或更有可能是在癌前病变进展期中起重要作用。Objective To investigate the mRNA expression of EphA2 in bladder tumor and its role in carcinogenic mechanism. Methods The expression of EphA2 mRNA were determined with real-time reverse transcriptase polymerase chain reaction（real-time RT-PCR） in 49 bladder urothelial carcinoma（BUC） , 19 inverted papilloma of the bladder（IPB） and 10 normal bladder tissues, and then the relative gene expression of EphA2 were studied by 2-△△CT method. Results Comparing to normal tissues, the expressions of EphA2 mRNA in IPB and BUC tissues increased respectively about 2.06（2.06±0.84） and 2.57（2.57±0.97） times on the average; There were no statistical differences of EphA2 mRNA expressions between IPB and BUC groups（P〉0. 05）; EphA2 mRNA expressions were positively correlated with the clinical stage（r=0. 471, P=0. 001） and pathological grade of BUC（r=0. 286, P=0. 047） and there were no correlations between EphA2 expressions and any other clinic and pathological features of BUC（P〉0.05）. Conclusions It was possible that EphA2 overexpressions facilitated IPB transforme into cancer as well as the malignant promotions of bladder tumor; the nucleic acid regulation mechanism of EphA2 may play an important role during the early formation and progress of bladder tumor.
