塞来昔布对Tca8113细胞侵袭性影响的体外研究

摘  要:目的:探讨环氧化酶-2(cyclooxygenease-2,COX-2)抑制剂——塞来昔布(celecoxib)对Tca8113细胞侵袭性的抑制作用。方法:用划痕实验检测Tca8113细胞的迁移能力;不同浓度的塞来昔布处理人舌鳞癌Tca8113细胞后,用MTT法检测细胞-基质黏附力的变化;用Transwell小室结合Matrix胶检测肿瘤细胞侵袭能力的变化。结果:10、20μmol/L的塞来昔布作用24 h后,细胞的侵袭能力分别下降了54%和73%(P<0.001),细胞-基质黏附力也明显降低(P<0.001)。结论:Tca8113细胞是高侵袭性细胞,塞来昔布对Tca8113细胞侵袭的抑制作用呈剂量依赖性。Objective:The purpose of this article was to investigate the effect of celecoxib(COX-2 inhibitor) on the aggressiveness of Tca8113 cell line.Methods:Traditional scape migration assay were used to examine the invasion of Tca8113 cells.After Tca8113 cells were treated with celecoxib,MTT assay was used to examine the adhesion between cancer cell and extracellular matrix.Transwell assay was used to examine the capability of invasion.Results:Celecoxib(10 and 20 μmol/L)significantly decreased the invasion ability of Tca8113 cells in vitro by 54% and 73% respectively(P 0.001).After the treatment of celecoxib for 24 h,the adhesion and invasion were decreased remarkably(P 0.001).Conclusion:Celecoxib may inhibit adhesion and invasion of Tca8113 cells in a dose-dependent manner to certain degree.
