摘 要:目的研究拳参正丁醇提取物(polygonum bistorla L.n-butylal cohol extract,PBNA)在大鼠视网膜缺血再灌注损伤时抗氧化能力及肿瘤坏死因子-α(tumor necrosis factorα,TNF-α)含量的影响。方法24只SD大鼠随机分成4组,即对照组、视网膜缺血再灌注模型组(简称模型组)、低剂量(0.3mg.kg-1)及高剂量(1.0mg.kg-1)PBNA组,每组各6只大鼠。颈总动脉夹闭法制作视网膜缺血再灌注模型。颈总动脉夹闭60min再灌注60min后,行视网膜电图(electroretinogram,ERG)检查;比色法测定血清丙二醛(malondialdehyde,MDA)、一氧化氮(nitricoxide,NO)浓度及超氧化物歧化酶(superoxide dismutase,SOD)、谷胱甘肽过氧化物酶(glutathione peroxidase,GSH-Px)活性,放射免疫法检测血清TNF-α含量。结果(1)与对照组ERG幅度(2.02±0.49)mV相比,模型组ERG幅度(0.55±0.28)mV显著降低,差异有显著统计学意义(P<0.01);与模型组比较,低剂量PBNA组ERG幅度(3.07±0.18)mV及高剂量PBNA组ERG幅度(3.71±0.48)mV均显著升高,差异均有显著统计学意义(均为P<0.001);(2)模型组血清MDA浓度(7.98±1.21)μmol.L-1较对照组(5.58±1.50)μmol.L-1升高,差异有统计学意义(P<0.05),与模型组比较,低剂量PBNA组MDA浓度(5.77±1.11)μmol.L-1降低,差异有统计学意义(P<0.05);(3)模型组血清NO浓度(26.31±5.34)mmol.L-1较对照组NO浓度(63.39±7.49)mmol.L-1降低,差异有显著统计学意义(P<0.001),与模型组比较,低剂量PBNA组NO含量(43.86±9.27)mmol.L-1升高,差异有统计学意义(P<0.05);(4)模型组血清GSH-Px活性(288.20±41.90)×103U.L-1较对照组(418.80±49.27)×103U.L-1降低,差异有显著统计学意义(P<0.01),与模型组比较,GSH-Px活性低剂量PBNA组(434.00±86.76)×103U.L-1及高剂量PBNA组(379.20±55.71)×103U.L-1均升高,前者差异有显著统计学意义(P<0.01),后者差异有统计学意义(P<0.05);(5)模型组血清SOD活性(64.19±7.65)×103U.L-1较对照组(77.56±7.76)×103U.L-1降低,差异有统计学意义(P<0.05),与模型组比�Objective To research the effect of polygonum bistorta L.n-butyl alcohol extract(PBNA) on anti-oxidation and the content of tumor necrosis factor-α(TNF-α) following retinal ischemia/reperfusion injury(RIRI).Methods Twenty-four SD rats were divided into control group,RIRI model group,low-dosage (0.3 mg·kg^-1) PBNA group and high-dosage (1.0 mg·kg^-1) PBNA group randomly.The RIRI model was established by dissociating and clamping the left common carotid artery.After clamped for 60 minutes and perfused for 60 minutes,the electroretinogram(ERG) were tested,the concentration of malondialdehyde(MDA),nitric oxide(NO) in serum and the activity of superoxide dismutase(SOD) and glutathione peroxidase(GSH-Px)were detected by colorimetric method,and the content of TNF-α by radioimmunology approach.Results (1)The amplitude of ERG in the control group,RIRI model group,low-dosage PBNA group and high-dosage PBNA group were(2.02±0.49)mv,(0.55±0.28)mv,(3.07±0.18)mv and (3.71±0.48)mv respectively,there were all significant differences between the RIRI model group and control group,low-dosage PBNA group,high-dosage PBNA group(all P〈0.01).(2)The concentration of MDA in the control group,RIRI model group and low-dosage PBNA group were (5.58±1.50)μmol·L^-1(7.98±1.21)μmol·L^-1and (5.77±1.11)μmol·L^-1respectively,there were all statistical differences between the RIRI model group and control group,low-dosage PBNA group(all P〈0.05).(3)The concentration of NO in the control group,RIRI model group,low-dosage PBNA group were (63.39±7.49)mmol·L^-1(26.31±5.34)mmol·L^-1and (43.86±9.27)mmol·L^-1respectively,there was significant difference between the control group and RIRI model group(P〈0.001),and statistical difference between the RIRI model group and low-dosage PBNA group(P〈0.05).(4)The activity of GSH-Px in the control group,RIRI model group,low-dosage PBNA group and high-dosage PBNA group were (41
