摘  要:目的观察视神经钳夹伤大鼠晶状体损伤对视网膜节细胞存活和轴突再生的影响。方法将32只SD大鼠随机分为正常组(8只)、对照组(8只)和处理组(16只)。正常组不作任何处理;对照组行单纯双眼视神经钳夹伤;处理组行视神经钳夹伤,同时双眼玻璃体注射5μL生理盐水,依据注射方式的不同分为晶状体损伤组和晶状体未损伤组。7d后荧光金逆行标记视网膜神经节细胞,14d后行视网膜铺片和节细胞计数,并用免疫组织化学法检测视网膜生长相关蛋白(growth associated pro-tein-43,GAP-43)和胶质纤维酸性蛋白(glial fibrillary acidic protein,GFAP)的表达。结果正常组和对照组的视网膜节细胞计数分别为(1987±154)mm-2和(908±123)mm-2,晶状体损伤组和晶状体未损伤组分别为(1206±134)mm-2和(800±112)mm-2。晶状体损伤组存活的节细胞数与对照组和晶状体未损伤组比较,差异均有统计学意义(均为P<0.05)。免疫组织化学法检测显示晶状体损伤组视网膜GAP-43和GFAP的表达亦比对照组和晶状体未损伤组强。结论晶状体损伤能促进视神经钳夹伤大鼠视网膜神经节细胞的存活,并增强视网膜GAP-43和GFAP的表达,促进轴突的再生。Objective To observe the effect of lens injury on retinal ganglion cells survival and axons regeneration in optic nerve crushed rats.Methods Thirty-two Sprague-Dawly(SD) rats were randomly divided into normal group,control group and treated group.Optic nerves of both eyes were crushed in control group.In treated group,5 μL normal saline was injected into vitreous body in both eyes after optic nerves were crushed,and then it was divided into two groups,one with lens injury and one without lens injury.For normal group,the above treatment was not done.Seven days later,the retinal ganglion cells were labeled with fluorogold was injected into both sides of superior colliculus of rats in each group.The number of retinal ganglion cells was determined in retinal whole mounts 14 days later.The expression of growth associated protein-43(GAP-43),glial fibrillary acidic protein(GFAP) were detected by immunohistochemistry.Results The number of retinal ganglion cells in normal group,control group,treated group with lens injury and without lens injury were (1 987±154)mm^-2,(908±123)mm^-2,(1 206±134)mm^-2 and (800±112)mm^-2.Compared the number of alive retinal ganglion cells between treated group with lens injury,control group and treated group without lens injury,there were all statistical differences(all P〈0.05).The results of immunohistochemistry showed that the expression of GAP-43 and GFAP in treated group with lens injury was greater than that in control group and treated group without lens injury.Conclusions Lens injury can increase the retinal ganglion cells survival in optic nerve crushed rats,boost the expression of GAP-43 and GFAP in retina and promote the regeneration of axons.
