生长抑素受体亚型2、5在胃癌细胞株中的表达及奥曲肽对胃癌细胞分泌VEGF的抑制作用

摘　　要：目的检测SGC-7901胃癌细胞株中生长抑素受体亚型2、5(SSTR-2、SSTR-5)的表达情况及其分泌血管内皮生长因子(VEGF)状况以及奥曲肽对SGC-7901胃癌细胞株分泌VEGF是否有抑制作用。方法采用免疫细胞化学及逆转录聚合酶链反应(RT-PCR)方法,检测胃癌细胞株SGC-7901中SSTR-2、SSTR-5的表达;利用酶联免疫吸附实验(ELISA)检测奥曲肽对胃癌细胞株SGC-7901分泌VEGF的抑制作用。结果SSTR-2、SSTR-5在胃癌细胞SGC-7901均呈阳性表达;SGC-7901胃癌细胞可以自分泌VEGF,呈时间依赖性;奥曲肽(10-6mol/L～10-5mol/L)可以抑制SGC-7901胃癌细胞合成分泌VEGF,具有时间-剂量依赖性。结论胃癌细胞株SGC-7901表达SSTR-2、SSTR-5;奥曲肽能够抑制人SGC-7901胃癌细胞中VEGF的分泌,可能是奥曲肽抗血管新生的作用机制之一。Objective To detect the expression of subtypes of somatostatin receptor （SSTR） and to investigate the effect of octreotide on production and secretion of vascular endothelial growth factor （VEGF） in SGC - 7901 gastric carcinoma cell line. Methods The expression of subtypes of somatostatin receptor （SSTR） was detected by using immunohistochemistry and RT -PCR methods. The effect of octreotide on VEGF secretion was measured by enzyme linked immunosorbant assay （ELISA）. Results Expression of SSTR - 2 and SSTR - 5 was detected in SGC - 7901 gastric cancer cell line. SGC -7901 gastric cancer cells could secrete VEGF. Octreotide in concentrations from 10^-6 mol/L- 10^-5 mol/L inhibited secretion of VEGF in a dose - dependent and time - dependent manner. Conclusion Subtypes ：2 and 5 of somatostatin receptor can be detected in gastric carcinoma and in SGC -7901 gastric cancer cell line. Octreotide is a potent inhibitor on production and secretion of VEGF in SGC -7901 gastric cancer cell line. It has been demonstrated that inhibitory action of octreotide on production and secretion of VEGF may be one of mechanisms underlying its anti - angiogenesis effect in SGC -7901 gastric cancer cells.
